One of the most interesting uses for cinnamic acid in recent years has been as a raw material in the preparation of L-phenylalanine [63-91-2] the key intermediate for the synthetic dipeptide sweetener aspartame (25). Genex has described a biosynthetic route to L-phenylalanine which involves treatment of immobilized ceUs of R rubra containing the enzyme phenylalanine ammonia lyase (PAT,) with ammonium cinnamate [25459-05-6] (26). [Pg.174]

American scientists prepared the organo-silica sol-gel membranes60 and demonstrated in a single layer format for pH measurement and multiple-layer format for both C02 and NH3. The sensors used a hydroxypyrenetrisulfonic acid (HPTS) as the indicator immobilizes in a base-catalyzed sol-gel containing poly(dimethyl)siloxane, aminopropyl-triethoxysilane (APTES) and tetraethylorthosilicate (TEOS). This indicator gel was over coated with a hydrophobic sol-gel to reduce cross reactivity to pH when either carbon dioxide or ammonia were examined. [Pg.367]

The members of Wolfbeis team constructed an optical sensor for ammonia-based on ion pairing76. They immobilized pH-sensitive dye (bromophenol blue) as an ion pair with cetyltrimethylammonium bromide (CTABr) in a silicone polymer matrix. Bromophenol blue, while contact the ammonia (both in water as well as in gaseous form) changes its color reversibly from yellow to blue. The immobilized dye shows long wave absorption with a good photostability. [Pg.370]

The same chemistry of binding has more recently been used for the immobilization of CDP-1 (see Section 2.1.2.7.5) via the two carboxylic acid groups present in the aglycone portion of the molecule, owing to the hydrolytic loss of ammonia [55]. [Pg.125]

Among potentiometric enzyme sensors, the urea enzyme electrode is the oldest (and the most important). The original version consisted of an enzyme layer immobilized in a polyacrylamide hydrophilic gel and fixed in a nylon netting attached to a Beckman 39137 glass electrode, sensitive to the alkali metal and NHj ions [19, 2A Because of the poor selectivity of this glass electrode, later versions contained a nonactin electrode [20,22] (cf. p. 187) and especially an ammonia gas probe [25] (cf. p. 72). This type of urea electrode is suitable for the determination of urea in blood and serum, at concentrations from 5 to 0.05 mM. Figure 8.2 shows the dependence of the electrode response... [Pg.202]

Polypropylene (PP) is a hydrophobic and chemically inert polymer which needs to be activated in order to be functional as a support for NA immobilization. Typically, PP membranes are aminated by exposure to an ammonia plasma generated by radiofrequency plasma discharge. Once aminated, the PP membranes can be reacted with derivatized ONDs using common coupling methods [56-58]. [Pg.94]

One of the pitfalls of microbial sensors, viz. their low selectivity, can be overcome by combining cells with an immobilized enzyme. Thus, creatinine deaminase (CDA, EC 3.5.4.21) hydrolyses creatinine to N-methylhydantoin and ammonium ion, the ammonia produced being successively oxidized to nitrite and nitrate ion by nitrifying bacteria. These bacteria have not yet been characterized but are known to be a mixed culture of Nitrosomonas sp. and Nitrobacter sp. The reaction sequence involved is as follows ... [Pg.128]

The bacteria consume oxygen during ammonia oxidation, so oxygen depletion can be detected by using an oxygen electrode. A combined creatinine sensor thus consists of a cellulose dialysis membrane, immobilized creatinine deaminase, immobilized nitrifying bacteria and an oxygen electrode [130]. [Pg.128]

Reflectance measurements provided an excellent means for building an ammonium ion sensor involving immobilization of a colorimetric acid-base indicator in the flow-cell depicted schematically in Fig. 3.38.C. The cell was furnished with a microporous PTFE membrane supported on the inner surface of the light window. The detection limit achieved was found to depend on the constant of the immobilized acid-base indicator used it was lO M for /7-Xylenol Blue (pAT, = 2.0). The response time was related to the ammonium ion concentration and ranged from 1 to 60 min. The sensor remained stable for over 6 months and was used to determine the analyte in real samples consisting of purified waste water, which was taken from a tank where the water was collected for release into the mimicipal waste water treatment plant. Since no significant interference fi-om acid compounds such as carbon dioxide or acetic acid was encountered, the sensor proved to be applicable to real samples after pH adjustment. The ammonium concentrations provided by the sensor were consistent with those obtained by ion chromatography, a spectrophotometric assay and an ammonia-selective electrode [269]. [Pg.184]

Isolated rat hepatocytes were immobilized in cellulose multiporous microcarriers by Kino et al. [24]. The microcarriers had a pore size of 100 pm and protected the cells from external shear stress. A newly developed stirred tank reactor contained the microcarrier-immobilized hepatocytes. The 02-supply was improved by using an oxygenator. The performance of microcarrier-im-mobihzed hepatocytes in the reactor was as good as that in floating culture and they demonstrated good ammonia metaboUsm. [Pg.104]
