HECK REACTIONS OF ARYL CHLORIDES CATALYZED BY PALLADIUM/TRI-tert-BUTYLPHOSPHINE (E)-2-METHYL-3-PHENYLACRYLIC ACID BUTYL ESTER AND (E)-4-(2-PHENYLETHENYL)BENZONITRILE... [Pg.32]

Pd/P(t-Bu)., in the presence of Cy2NMe, is an unusually mild and versatile catalyst for Heck reactions of aryl chlorides (Tables 1 and 2) (as well as for room-temperature reactions of aryl bromides).21 22 23 Example A, the coupling of chlorobenzene with butyl methacrylate, illustrates the application of this method to the stereoselective synthesis of a trisubstituted olefin a-methylcinnamic acid derivatives are an important family of compounds that possess biological activity (e.g., hypolipidemic24 and antibiotic25) and serve as intermediates in the synthesis of pharmaceuticals (e.g., Sulindac, a non-steroidal anti-inflammatory drug26). Example B, the coupling of 4-chlorobenzonitrile with styrene, demonstrates that Pd/P(t-Bu). can catalyze the Heck reaction of activated aryl chlorides at room temperature. [Pg.35]

Dominguez and coworkers [41] used a twofold Heck reaction for the construction of annulated N-heterocycles such as 6/l-64a-c, starting from the enamides 6/1-63 which can be easily obtained from the corresponding amines 6/l-62a-c and o-iodo-benzoic acid chloride (Scheme 6/1.15). [Pg.369]

Synthesis of dithieno[2,3-A2,3-,7]thiophene derivatives 122 has been accomplished through the Heck reaction of 3-(4-bromo-2-thienyl)acrylic acid 302 to afford 3-(2,4-thienylene)diacrylic acid 303 which was cyclized with thionyl chloride and a catalytic amount of pyridine to the dichloride 120 in 75% yield (Scheme 56) <2005MOL279>. [Pg.674]

Several groups intercepted the indole-palladium complex that is initially obtained on cyclization by a subsequent Heck reaction. As will be seen, this can be a powerful elaboration of indoles. In the first example of this concept, Utimoto and co-workers ambushed intermediate 354 with a series of allylic chlorides to give 355. Normal acid workup yields the corresponding C-3 unsubstituted indoles (52-83%) [310]. [Pg.155]

Scheme 2 shows a similar mechanistic pathway for a Heck reaction taking place on a Pd octahedral comer. This mechanism is based on that established for soluble Pd catalysts (ref. 5). Adsorption of the aryl halide (or aryl acid chloride after decarbonylation) gives the aryl Pd halide, 15, by way of the adsorbed intermediate, 14. Vinyl ether adsorption, as in 16, takes place as described in Scheme 1. Aryl insertion gives the halometalalkyl, 17, which on f) elimination to the available 4dxy orbital gives the aryl enol ether, 2 (or 1 depending on which hydrogen is eliminated in 17). The resulting halo palladium hydride, 18, then reacts with the tertiary amine to give the amine hydrochloride and regenerates the octahedral comer for further reaction. C-C coupling reactions include Pd-catalyzed carbonylations as well as Heck reactions. Biphasic operations are reported as being particularly efficient. Allyl and benzyl chlorides have been converted in two-phase systems to the corresponding acids with or without phase-transfer catalysts (eq. (6)) [154-157, 237]. [Pg.618]

Blaser and Spencer used aroyl halides in place of aryl halides, with aroyl chlorides being of specific interest as ubiquitous, relatively cheap compounds ( Blaser reaction ) [24], This latter reaction is normally conducted in aromatic solvents phosphines are not used here as catalyst ligands since they fully inhibit the reaction. In the same way, benzoic acid anhydrides can be used as the aryl source in combination with PdCl2 and catalytic amounts of NaBr [79]. In this reaction, one of the arenes is used in the coupling reaction by elimination of CO, whereas the other benzoate serves as the base. The benzoic acid thus formed can easily be recycled into the anhydride. The use of aryl and vinyl triflates according to Cacchi [25] and Stille [26] extends the scope of the Heck coupling to carbonyl compounds phenol derivatives act via triflate functionalization as synthetic equivalents of the aryl halides. The arylation of cyclic alkenes [27], electron-rich vinyl ethers [28], and allylic alcohols [29] is accessible through Heck reactions. Allylic alcohols yield C-C-saturated carbonyl compounds (aldehydes) for mechanistic reasons (y9-H elimination), as exemplified in eq. (6). [Pg.779]

As mentioned in the discussion of the pathways to indoles (Scheme 27), a detailed indole synthesis with two points of diversity based on the Heck reaction has been reported [164]. The indole core structure was synthesized via a 5-exo-tng transition state, which provided the exocyclic double bond that subsequently underwent exo to endo double-bond migration. The anthranilate building block was prepared in solution and immobilized by a method previously described for the loading of 2-aminobenzophenones [Ij. After Fmoc cleavage, the resulting 4-bromo-3-amino-phenyl ether was treated with acid chlorides and pyridine in CH2CI2. As outlined in Scheme 29, alkylation of the anilide with substituted allyl bromides was achieved in the presence of lithium benzyloxazoHdinone in THF. The reaction mixture was treated with base for 1 h and then an aUylic halide was added and the mixture was vortexed for 6 h at room temperature. The alkylation reactions were... [Pg.424]

An example is as follows. After protection as a silyl ether, amidation of benzyl A-(5,6-0-isopropylidene-4-hydroxycyclohex-2-enyl)carbamate 229 (prepared from 3-bromo-lS,2S-0-isopropylidenecyclohexa-3,5-diene) with (6-bromo-3,4-methylenedioxy)benzoyl chloride gave the protected N-benzyloxycarbonyl-A-(cyclohex-2-enyl)benzamide 230. A modified Heck reaction of 230 in anisole produced the protected lycoricidine 231 in 27% yield (two steps) by A-detosylation. Acid treatment of 231 provided (-H)-lycoricidine (204) in 85% yield (Scheme 22). [Pg.373]
