Sprinklers and fire protection systems are required by NFPA Codes, but are often dependent on the overall size of the facility and quantity of stored flammable/combustible material. The wisest course of action is to provide heat and smoke detectors in each laboratory and provide a sprinkler system at least in the hallways. Each laboratory should have at least one ABC portable fire extinguisher. Computers have become more important to laboratories than ever. Halon fire extinguishing systems are available which are nondestructive to both electronic equipment and human life. These should be employed for fire protection. [Pg.230]

Whereas antigen-retrieval technique serves to amplifying the immunocytochemical signal at the predetection phase, conventional methods of signal amplification, such as avidin biotin complex (ABC) and soluble enzyme-anti-enzyme immune complex techniques (peroxidase-anti-peroxidase complex and alkaline phosphatase-anti-alkaline phosphatase complex PAP and APAAP respectively), are applied in the phase of detection. For many years, the PAP and APAAP procedures represented the most sensitive and reliable and hence most popular techniques in many pathology laboratories. However, today these techniques are only rarely used, being substituted by modem more sensitive methods. [Pg.51]

Avidin-biotin complex horseradish peroxidase reagent (Vectastain ABC HRP Kit, Vector Laboratories, Inc., Burlingame, CA). [Pg.206]

Prepare the ABC reagent according to the manufacturer s instructions. For Vector Laboratories Vectastain Elite kit, add 2 drops of reagent A to 5 mL of PBS, then add 2 drops of reagent B. Mix and incubate for 30 min at room temperature. Additional volume may be prepared keeping the ratio of reagents constant. Repeat step 6. [Pg.207]

Avidin-biotin complex (Vector ABC Elite kit) (Vector Laboratories) ABC solution should be made fresh 10 min before use, so that complexes have time to form according to instractions of manufacturer. [Pg.216]

The sections are incubated for 72 hr at 4°C with primary antiserum in PBS containing 2% normal serum and 0.25% gum arabic (Sigma). The sections are then incubated overnight at 4°C with a biotinylated secondary antibody (Vector Laboratories, Burlingame, CA), diluted 1 200. They are next incubated overnight at4°C with avidin-biotinylated peroxidase complex (ABC) diluted 1 100 in PBS containing 0.25% gum arabic. Note that the peroxidase-antiperoxidase procedure can also be used. [Pg.178]

The sections are rinsed in the same buffer and incubated for 90 min in secondary antiserum (Jackson ImmunoResearch Laboratories, West Grove, PA), diluted 1 50 with PBX-5% normal horse serum. They are rinsed in 0.1 M sodium phosphate buffer and then incubated for 1 hr in peroxidase antiperoxidase (PAP), diluted 1 100 with PBX. The sections are rinsed three times for 5 min each in the same buffer followed by distilled water rinses. They are incubated for 10 min in 50 ml of 0.05 M imidazole/0.05 M cacodylate buffer (pH 7.2) containing 50 mg of DAB. This is followed by an additional 10 min incubation after adding 200 xl H202. All incubations are carried out under constant agitation. The sections are washed in distilled water, placed in the same buffer, mounted onto gelatin-coated slides, dried, dehydrated, and cover-slipped with Permount. Note that the ABC procedure can be used instead of the PAP procedure. [Pg.180]

Endogenous peroxidase activity is quenched by treatment with 0.5% H202 in methanol, immunostaining is carried out using a Vectastain Elite ABC kit, and visualization of the secondary antibody is achieved with DAB enhanced with nickel (Vector Laboratories). [Pg.192]

TABLE 3.1 ABC Water Laboratory Activities, Manipulatives, and Real-World Connections... [Pg.57]

The ABC units answer the so what question by providing contexts that students can relate to. In the Water unit, for example, a laboratory entitled How Is pH Lowered begins by introducing a fictional character who lowers pH as part of her job ... [Pg.57]

The ABC materials encourage transfer in numerous instances. Particularly good examples are found in the Challenge sections that conclude the laboratories. The Water unit, for example, includes a laboratory designed to help students understand how molecules move across membranes. The stated purpose of the laboratory is to help students determine the effect of concentration difference on the movement of water and solute across a membrane. The laboratory s stated objective is to enable students to predict the direction of material movement across a membrane based on the concentration of materials on both sides of the membrane. During the laboratory, students measure mass with a balance and work with dialysis bags. At the conclusion of the laboratory, students explore questions designed to help them transfer what they have learned to contexts outside the classroom ... [Pg.60]
