Another radionuclide that has been commonly used in physiological studies of terrestrial animals is tritium. As stated in Section 8.2.1, water can be labeled as HTO and the compound used to study the water pool in plants. Somewhat similarly, total body water and water turnover in mammals can be studied. Longhurst et aL (1970) utilized HTO to compare water kinetics in winter and summer Columbian black-tailed deer (Odocoileus hemionus columbianus) and domestic sheet (Ovis aries). The animals were injected intravenously with 8 mCi HTO, and blood samples were collected periodically and examined for radioactivity with a liquid scintillation counting system. In a water-turnover study in mule deer (Odocoileus hemionus), Knox et aL (1969) also used HTO and observed that the biological half-life of HTO differed in animals maintained in a 0.9 X 1.5 m metabolism stall from that in the same animals maintained in a 4.6 X 5.8 m room. [Pg.120]

Two linear columns from Showa Denko, Shodex SB-806M and Shodex SB-806MHQ, and two linear columns from TosoHaas, TSK GM-PWxl and TSK GM-PW, were evaluated. Prior to the evaluation, the number of theoretical plates for Shodex SB-806MHQ, SB-806M, PWxl, and PW was determined to be 15,100, 15,700, 11,390, and 4710, respectively, as per manufacturer inspection. The lower plate count of the TSK PW column is due to the larger particle size of this column. Two mobile phases, water with 0.1 M LiNOi and 50 50 methanol/water (v/v) with 0.1 M LiNOi, were used for each of the four columns. These four columns were new and only PEO and PVP were analyzed with these columns in this study. Waters Ultrahydrogel columns have also been used in this laboratory. The metabolic and pharmacokinetic profile of sucralose (this is a novel intense sweetener with a potency about 600 times that of sucrose) in human volunteers was studied by Roberts and coworkers [82]. Part of this study was realized using PLC in the following chromatographic system in which the stationary phase was silica gel and the mobile phase was ethyl acetate-methanol-water-concentrated ammonia (60 20 10 2, v/v). Separated substances were scraped off separately, suspended in methanol, and analyzed by filtration, scintillation counting, or enzymatic assay. Procedure Pollen develops on the nutrition medium, forming pollen tube (Fig. 3). The nutrient medium for pollen was 10% sucrose solution and tested compounds were also dissolved in 10% sucrose. The pollen develops till the formation of pollen tube that lasts from 2-3 h (fresh collected and one week stored pollen) to 24 h (stored > 1 week, but < 1.5-2 months). All experiments were done at room temperature 20-22 °C. The growth occurred in the solution studied (0.05 ml = 1 drop) on the slides (object glasses) put on wet paper in Petri dishes. Five ml of water was added to the bottom of every dish and 4-5 dishes with the slides were used per treatment. Using light microscope, we determined the microspores germination (%) 2-24 h after moistening. The number of developed pollen tubes was counted. [Pg.33]
