The analysis of amino acids involves chromatographic issues similar to those encountered in analysis of simple amines. Underivatized amino acids have, with a few exceptions, weak UV absorbance and a strong tendency to interact with stationary phases in undesirable ways. Underivatized amino acids are normally separated with ion exchange chromatography, then visualized post-column by reaction with ninhydrin, o-phthaladehyde (OPA), or other agents. For PK assays, it is generally believed that most matrix effects are due to the sample matrix (typically plasma). While this is correct in many cases, this assumption has some exceptions (vide infra). One of the most useful tools for avoiding matrix effects is studying the sample matrix and proposed assay by using the post-column infusion technique described by Bonfiglio et al.14S This technique allows visualization of the portion of the chromatographic step affected by ion suppression.161721 Xu et al.101 recommended inclusion of this step in the method development process for drug discovery PK assays. [Pg.220]

Strength Habitability surveys conducted at CP (command post) upon arrival of Haz-Mat team. EPA conducted surveys at CP upon their arrival. SRS (Savannah River Site) and Richmond County Haz-Mat resources arrived on scene within a timely manner and were designated by FDIC (Fire Department Incident Commander) to be responsible for Haz-Mat operations. Written response plan and safety procedures implemented for Haz-Mat operations. The concentration of carbon tetrachloride was measured in the expired air of a person who swallowed a large amount of carbon tetrachloride (Stewart et al. 1963). Excretion in expired air was found to decrease exponentially in a biphasic or multiphasic fashion, but no quantitative estimate of the elimination half-life of carbon tetrachloride or of the fraction of the dose excreted by this pathway was provided. Visual inspection of their graphed data suggests very approximate half-lives of less than several hours initially, 40 hours (75-150 hours post exposure), and 85 hours (300-400 hours post exposure). [Pg.71]

The mechanisms of flashbacks are probably mixed. Some cases may be similar to post-traumatic stress disorder induced by a bad trip (Paton et al., 1973). Abraham (1983) suggested that some of the visual phenomena, such as trailing and after-images, were due to failure of inhibition in visual pathways, possibly mediated in the lateral geniculate nucleus which (in the macaque monkey) contains on-off colour neurons with receptor fields similar to those described in flashbacks. The neurochemical causes of such flashbacks, which can be very disturbing, remains elusive and attempts at treatment are usually ineffective. [Pg.198]
