The cross-section of the primary X-ray beam is extended and not an ideal point. This fact results in a blurring of the recorded scattering pattern. By keeping the cross-section tiny, modern equipment is close to the point-focus collimation approximation - because, in general, the features of the scattering patterns are relatively broad. Care must be taken, if narrow peaks like equatorial streaks (cf. p. 166) are observed and discussed. The solution is either to desmear the scattering pattern or to correct the determined structure parameters for the integral breadth of the beam profile (Sect. 9.7). [Pg.56]

For over 300 years, the quinoline family of drugs, and chloroquine in particular, has been used as the primary treatment for malaria. Recent studies have demonstrated that this drug inhibits the aggregation of free heme into hemozoin, allowing levels of monomeric heme to rise until cell lysis occurs. Although the determined structure of hemozoin makes the polymer termination scheme proposed by Sullivan et al. unlikely, hemozoin drug heme interactions appear critical in the inhibition mechanism. Thus, researchers have identified the characteristics of... [Pg.360]

Whatever approach is used to deal with the phase problem, the final result will be a pattern of electron density in the unit cell which is then fitted to the amino add sequence of the molecule, which is usually known, to produce a three-dimensional structure. From this structure, a calculated diffraction pattern can be evaluated for comparison with the experimentally observed pattern. A measure of the quality of the determined structure is given by the so-called R factor which is defined by the following equation ... [Pg.290]

An estimation of the TST rate constants was performed from the determined structures, energies, and vibrational modes of all of the reaction components. These constants are collected for both directions of the reactions (k, k i, k2, and k 2) at temperature 310 K in Table 7. Comparing the calculated constants for the dechlorination process in the 1st hydration step with the available experimental values,25-39 124 a qualitative agreement has been reached. It is clear that differences of several orders of magnitude in the values of the rate constants represent a variation of only a few kcal/mol on the hydration surface. Hence, the requirement for accuracy for such kinetic quantity is at the limit of the current computational techniques for the... [Pg.305]

In addition to the two examples described above, IR spectrometric methods have been applied to a few proteins, such as bovine serum albumin (BS A) (14,15) or lysozyme (16). They provided hydration mechanisms similar to those described above, but the relation between the determined structure of the H-bond network at a definite hygrometry and properties of... [Pg.263]

Structural parameters for transition metal adsorption on transition metals epitaxial structures. The given bond length is the shortest distance between the adatom and a substrate atom. du is the spacing between the first and second layers of the epitaxial system, di3 is the spacing between the second and third layers, etc. The adatom-adatom and adatom-substrate bond lengths are derived from the determined structural paramaters. [Pg.30]

The alignment site 228 in Fig. 10(b) is occupied by a His residue that was previously considered to act as a ligand for the haem iron (for example, see review [87]). However, the determined structure of ovine COX-1 revealed that the residue His-309 in the enyme does not bind to the haem group, but constitutes an interface between helices H5 and H6. As shown in Fig. 10(b), helices H5 and H6 are present at a highly conserved region among COXs and peroxidases. In the structures of both ovine COX-1 and canine myeloperoxidase, the two helices form a V -shaped core structure of the large lobe, around which helices H3, HIO, H18 and H19 are... [Pg.39]

As tissue ages, these initial cross-links decline in quantity as they are converted into other, more complex mature cross-links. These multifunctional cross-links are key to providing strength and stability to tissues. Despite their importance, however, the determined structures are limited (Fig. 6) and other structures may also be present. For some cross-links, such as histidinohydroxylysinonorleucine and pyridinoline, the structures are well defined and the paths to their formation well imderstood (Fig. 6). For others, the information is not as complete. The types and extent of cross-links vary between tissues in a specific manner (50). Finally, in addition to these specific cross-links, as tissue ages it may accumulate a range of additional cross-links that form because of nonenzymatic glycosylation of collagen (51). [Pg.1518]
