Lysis, cell

The first step in the extraction of nucleic acids may require the lysis of cells and the inactivation of cellular nucleases. These two steps may be combined into one. For instance, a single solution may contain detergents to solubilize the cell membrane and strong chaotropic salts to inactivate the intracellular [Pg.333]

There are several methods of cell lysis (Table 8.2) [1,2], but there is none that works with cells of all biological origins. Each technique has its advantages and disadvantages, and the specific method of choice depends on the cell characteristics, the cell type, and the final application. A combination of more than one method may also be used. For example, enzymatic lysis uses specific enzymes to target the cell wall. However, to disrupt the cytoplasmic [Pg.333]

The mode of action of starch capped copper nanoparticles (SCuNPs) was compared with that of the well-known antibiotic amphicillin (Fig. 9). Patients with tumor lysis syndrome experience a wide range of metabolic abnormalities. The massive cell lysis that occurs leads to the release of intracellular electrolytes, resulting in hyperkalemia and hyperphosphatemia. Molecular methods used to uncover mutations are subject to several variables. The anticoagulants used for blood collection can affect digestion with restriction enzymes and amplification reactions. The type of detergent used in cell lysis can affect amplification of DNA by inhibiting the DNA-amplifying enzyme such as the taq polymerase used in the polymerase chain reaction (116). One note of caution with regard to these types of studies is that one must take into account any other cellular mechanisms for substrate and/or product depletion. For example, in studies of enzymes that act on protein substrates, one must ensure that the substrate and products are isolated from cells under conditions that do not promote their destruction. In the case of kinases, for example, one must ensure that cellular phosphatases are inhibited during cell lysis and sample preparation, to ensure that substrate buildup is not the result of phosphatase-catalyzed dephosphorylation of the kinase product. [Pg.137]
