T cell repertoire

Pepin et al. (1992) generated transgenic mice in which antisense RNA complementary to GR cDNA led to reduced expression mostly in neuronal tissues. Consequently, this was found to result in an impaired behavior, a defective response to stress as well as in obesity. King et al. (1995) generated transgenic mice where reduced GR expression was limited to the thymus. This leads to an altered thymocyte development, changes in the T-cell repertoire, and a reduced risk to develop autoimmune diseases. [Pg.546]

Although central tolerance is the major mechanism to establish the T-cell repertoire by positive and negative selection mechanisms, thymic deletion of harmful T-cell populations is incomplete. Therefore, the immune system has developed mechanisms that deal with tolerance in the peripheral lymphoid organs providing the necessary... [Pg.179]

In conclusion, these results demonstrate that affinity and specificity of A2.1-restricted TCRs, selected in Tg mice by circumventing self-tolerance to universal hdm2- and wt p53-derived CTL epitopes, can be successfully delivered into human T lymphocytes. This, however, is precisely the molecular requirement to rescue the human T cell repertoire with high-affinity leukemia and tumor-reactive TCRs that have been lost due to the establishment of antigen-specific self-tolerance [13, 14]. [Pg.249]

HIV-specific CD8+ function may result from continued mutation and high levels of viral turnover, which gradually diminishes the capability of the cell to recognize the genetic sequences of the virus due to killing of some of the CD8 T cells repertoire by HIV itself. [Pg.175]

The immune windows , along with developmental windows for the respiratory system, were identified at a workshop sponsored by the USEPA and the March of Dimes (see Dietert et al., 2000 Holladay Smialowicz, 2000 Landreth, 2002). They include 1) initiation of haematopoiesis, 2) migration and expansion of stem cell populations, 3) colonization events, including bone marrow colonization, pre-T cell seeding to the thymus, T cell education, T cell repertoire establishment, then seeding of the periphery by mature T cells, 4) acquisition of immunocompetence, and, finally, 5) the... [Pg.98]

The demonstration that SAgs encoded by endogenous murine retroviruses caused the deletion of CD4+CD8+ precursors in the thymus provided the first formal evidence for a negative selection of the T cell repertoire based on clonal deletion [16, 17]. It was subsequently shown that the bSAg SEB also caused deletion of CD4+CD8+ precursors in the thymus [18]. [Pg.139]
