Supplementary procedures

The roller bottle system is a multiple process requiring considerable staff time for repeated manipulations. Thus many modifications have been introduced that increase the efficiency or surface area capacity of roller bottles. Some examples are as follows  

Perfusion. Specially modified swivel caps have been developed that allow continuous perfusion of roller bottles (e.g. Bellco Autoharvester, New Brunswick apparatus). 

Spira-Cell Multi-Surface Roller Bottle (Bibby Sterihn Ltd). This bottle contains a spiral polystyrene cartridge on which cells grow on both sides of the film 3000-cm, 4500-cm and bOOO-cm sizes are available. 

Extended Surface Area Roller Bottle (ESRB) (Bibby Sterilin Ltd). This polystyrene bottle has a ribbed or corrugated surface, doubling the surface area of the SSO-cm flask to 1700 cm.  

Bellco-Corbeil Culture System (Bellco). A roller bottle is packed with a cluster of small glass tubes (arranged in parallel and separated by silicone spacer rings). Models are available in lOOO-cm, 10 OOO-cm and 15 000-cm sizes. Medium is perfused through the vessel and the bottles rotate 360° in alternate directions to avoid tube twisting and the need for special caps.

Managing electronic records and electronic signatures may be handled with separate SOPs or integrated into the above. An abridged set of procedures will be appropriate for small systems, but supplementary procedures will be needed for larger ones. 

For studies involving metabohsm-mediated effects of xenobiotics, cells that are known to contain the requisite mixed-function oxygenases should be selected. As an alternative, an S9 hepatic microsomal fraction (see Supplementary procedure bioactivation) can be included in the test medium (Borenfreund Puerner, 1987). Serum concentrations should be kept as low as tolerated (or eliminated as in a defined medium) to prevent binding of the test agent to serum components.
