Sulphonamides were supplied by Sigma (Sigma Chemical Company, St. Louis, MA 63178 USA) Sulphisomidine (SOMI), Sulphathiazole (THIA), Sulphamethizole (METH), Phtalylsulphacetamide (FTAL), Sulphacetamide (ACET), Sulphapyridazine (PYRI), Sulphamerazine (MERA), Sulphamethoxypyridazine (MEPY) and Sulphachloropyridazine (CLPY). 

Figure 7.9 gives the minimal partition coefficient plot of the extraction of sulphapyridazine, sulphamerazine, sulphamethox)q)yridazine and sulphachlorop)ridazine. 

It can be seen from this figure that the minimal partition coefficient plot is dominated by two of the four compounds, namely by sulphamerazine in the methylene chloride region and in the methyl tert.-butyl ether region and by sulphapyridazine in the chloroform region.

In Hb Zurich, the E 7 (63 fi) distal histidine is replaced by arginine at the same site the replacement by tyrosine in Hb M Saskatoon favours the oxidation to methaemoglobin. The following drugs inducing haemolytic reactions in G-6-PD deficiency are also found to cause haemolysis of red cells which contain Hb Zurich sulphisoxazole, sulphisomidine, sulphadimethoxine. sulphamerazine. sulphamethoxy-pyridazine, primaquine and other oxyquinolines. 

It is perhaps relevant to note here that in clinical use sulphonamide mixtures are preferred to single drugs, to minimize the formation of crystal deposition in the kidney. The use of mixtures of related sulphonamides results in enhanced mutual solubility [284]. This is perhaps a case of hydrotropy. Lehr [285] has determined the solubility of a number of sulphonamide combinations, and Frisk et al. [286] have reported on the solubility of the combination sulphadiazine, sulphamerazine, and sulphathiazole. The influence of sulphanilamide on the solubility of sulphathiazole indicated that a 1 1 molecular complex was formed [287].

The degree of flocculation of sulphamerazine suspensions has been determined as a function of surfactant (sodium dod yl polyoxyethylene sulphate) concentration and electrolyte concentration [14]. At low surfactant levels the system is flocculated, with sedimentation volume fractions of about 0.9 at higher levels the sedimentation volume falls to about 0.6. On the addition of 0.24 m NaCl a dramatic decrease in sedimentation volume is observed, indicating a dense sediment with cloudy supernatant. Further addition of NaCl results in reestablishment of a flocculated state perhaps by adsorption of ions on to the particles.
