APPENDIX REAL-TIME METHODS FOR ANALYSIS OF LIGAND BINDING Method 1. Spectrofluorometric Method Using Antibody to Fluorescein... [Pg.65]

Spectrofluorometric methods of analysis are the most commonly analytical techniques and continue to enjoy wide popularity. The wide availability of the instrumentation, the simplicity of procedure, sensitivity, selectivity, precision, accuracy, and speed of the technique still make the spectrofluorometric methods attractive. These features make fluorescence spectroscopy an attractive technique as compared to other forms of optical spectroscopy or other analytical techniques such as chromatography and electrophoresis. Fluorescence spectroscopy has been used widely as a tool for quantitative analysis, characterization, and quality control in the pharmaceutical, environmental, agricultural, nanotechnology and biomedical fields. [Pg.209]

Highly sensitive spectrofluorometric measurements have been used extensively in pentazocine analysis especially in measurements from biological sources. The method developed early by Berkowitz, Way and coworkers (27,4-9,50,51.52) has been modified and extended by other investigators (53.54-, 55,56). Pentazocine levels in plasma, urine, brain, and intestine have been determined by this method. Briefly the tissue or fluid is basified with a... [Pg.389]

In the fluorometric method I, the NADH produced is monitored spectrofluorometrically at an excitation wavelength (Xex nm and an emission wavelength ) of 467 nm. The fluorescence intensity is proportional to the concentration of formaldehyde. Alternatively, the following coupled reaction can be used for more sensitive analysis of formaldehyde in the ppb concentrations ... [Pg.118]

Methods of analysis. The total protein concentration of crude solutions, including cellular proteins, was determin by the biuret method (72). With dilute xanthan solutions, this method was not sensitive enough, and a spectrofluorometric assay was carried out. This method consisted of an acid hydrolysis of the samples followed by derivatization with o-phthaldialdehyde and HPLC analysis on a Cl8 reverse column of the amino acids released, using spectrofiuorescence as the detection nKxie. [Pg.255]

A unique stopped-FIA system for simultaneous synchronous spectrofluorometric online dissolution monitoring of multicomponent solid preparations that makes use of a fiberoptic sensor was presented by Li et al. [3]. A new means of in vitro therapeutic drug monitoring was developed by hyphenating the fiberoptic sensor technique and a chemometric method. An artificial neural network method was applied to construct the mathematical model for the simultaneous analysis of the mixture of vitamins Bj, B2, and Bg by synchronous spectrofluorometry. The selection of the wavelength interval, the pFI of the carrier solution, and other experimental conditions were evaluated. The proposed method has been applied to the dissolution monitoring of vitamin B tablets with satisfactory results. The recovery was 97.8%-105%, and the relative standard deviation (RSD) was 1.1-7.5. [Pg.490]
