Snips

Skin snips may be useful in the diagnosis of microfilarial infections such as onchocerciasis in which the parasites circulate in the skin and not the blood. A small (2-mm) skin snip is taken with a needle and a knife. The needle point is stuck into the skin, and the skin is raised. With a sharp knife or razor blade, the skin is excised just below the needle. Alternatively, a scleral punch may be used. The skin snip is then placed in a small volume (0.2 ml) of saline in a tube or a microtiter well, teased, and allowed to stand for 30 min or more. The microfilariae migrate from the tissue into the saline, which is then examined microscopically to demonstrate the wiggling microfilariae. 

Detach the ends of these two wires from any others to which connected and strip the insulation from the last half inch or so. The other wires may be snipped off or just folded out of the way. 

Suppose t(P) is not liberal and the smallest "illiberal" execution sequence has length m. If we examine the tree scheme corresponding to paths in t(P) of length less than m this must be free. Hence if this execution sequence has a subpath passing twice through the same address we can "snip" out the subpath and still have a legitimate execution sequence.

Take the opportunity when watering to look for pests and diseases, and snip off any infested foliage. 

Pick sweet cherries as soon as they are ripe either eat them immediately, refrigerate for a short time, or freeze. Sour cherries do not pull away easily from the tree and may need to be snipped off. Fruit is ready when very dark in color and soft to the touch. It can be used to make jam or wine, preserved in brandy, or cooked for immediate use or freezing. 

For most reactions, one can use 1-2 mM acetyl phosphate and 1-2 international units of acetate kinase. The enzyme is usually supplied as a crystalhne suspension in 3 M ammonium sulfate, and 10 mM ammonium ion is inhibitory. Therefore, a useful practice is to snip off 0.5 cm from a disposable Eppendorf micropipette tip to facih-tate removal of 10-20 microliters of the crystalline suspension then spin down the enzyme in a 1.5 ml disposable conical plastic centrifuge tube, and remove the ammonium sulfate solution with a wick of twisted Kim-wipe. The enzyme precipitate can now be taken up directly into your working buffer. Note Acetate kinase is inactivated by cold exposure, but incubation with 10 M ATP or GTP reactivates the enzyme if warmed to room temperature for 5-10 min.
