Serum albumin conjugates fluorescence

Weber G (1952) Polarization of the fluorescence of macromolecules II. Fluorescent conjugates of ovalbumin and bovine serum albumin. Biochem J 51 155-167... [Pg.59]

When staining with any of the fluorescent phalloidins, dilute 10 pL methanolic stock solution into 400 pi PBS. To reduce nonspecific background staining with these conjugates, add 1% bovine serum albumin (BSA) to the staining solution. It may also be useful to preincubate fixed cells with PBS containing 1 % BSA. [Pg.93]

Secondary antibody usually fluorescent antimouse conjugate, stock solution diluted 1 20-1 500 in PBSG with 10% bovine serum albumin see Note 1). [Pg.262]

Finally, a model protein bioconjugate 52 was prepared with the reactive fluorophore 50 and bovine serum albumin (BSA). The conjugate was identified spectrophotometrically and its steady state fluorescence emission spectra... [Pg.135]

In 2001, Van Orden et al. reported the first biologically relevant FRET investigation using QDs as energy donors.83 They conjugated biotinylated bovine serum albumin (bBSA) to water-soluble CdSe-ZnS-TA QDs via a thiol linkage with 2-iminothiolane. The acceptor was streptavidin covalently labeled with tetra-methylrhodamine (TMR). Once the avidin-biotin interaction took place, a decrease in the QD luminescence and an increase in the TMR luminescence were observed, which was attributed to FRET between QD and TMR. Although no time-resolved fluorescence experiments were carried out and no estimates for the separation distance... [Pg.390]

Acrolein-containing latex particles were used for immobilization of proteins. The latex particles were prepared by emulsifier-free polymerization of acrolein and styrene (Table I). Fluorescent latex particles were prepared by mixing fluorescent dyes such as Hostalux KCB (Hoechst) or coumarin-6 into styrene before the emulsion polymerization (Table I). To increase the immunological sensitivity of the protein-conjugated latex particles, a hexyl group was introduced between the latex particle and the protein (Protein-spacer-latex). Proteins such as human serum albumin (HSA), anti-human serum albumin (anti-HSA-IgG), and a fragmented antibody (anti-HSA-... [Pg.285]

Figure 12.7 Protein-protein interaction assays performed on filled-squared ZnO NR arrays. (A) Interaction between biotinylated bovine serum albumin (BBSA) and dichlorotriazinylaminofluorescein (DTAF) conjugated streptavidin leads to strong fluorescence emission shown in panels 4 and 5. As-grown ZnO NR arrays do not show any autofluorescence. SEM micrograph of as-synthesized NR arrays is displayed in panel 1 and the corresponding fluorescence emission of as-grown NR arrays is shown in panel 2. Ever since the demonstration that synthetic peptides could target a protein to the nucleus when cross-linked to it (Goldfarb et ai, 1986), peptide-protein conjugates have been used widely in nuclear protein import assays in vivo and in vitro. The method described here is for the preparation of a fluorescent bovine serum albumin, SV40 NLS conjugate. However, this basic protocol can be adapted easily to allow the conjugation to other proteins, such as the naturally fluorescent protein allophycocyanin (Adam etai, 1991). [Pg.523]

Transfer from nanoparticle donors to tetramethylrhodamine (TMR) acceptors This was described for the binding between biotinylated bovine serum albumin (BSA) conjugate with ZnS(CdSe) nanoparticles and TMR-labeled strepta-vidin by observing enhanced TMR fluorescence. [Pg.30]
