Bromelain (anti-inflammatory Ananase from pineapple) [37189-34-7] Mr 33 000, [EC 3.4.33.4]. This protease has been purified via the acetone powder, G-75 Sephadex gel filtration and Bio-Rex 70 ion-exchange chromatography and has Aj 20.1 at 280nm. The protease from pineapple hydrolyses benzoyl glycine ethyl ester with a Km (app) of 210mM and kcat of 0.36 sec. Figure 3 shows the final chromatogram and activity profile of purified alpha-endopsychosin. The first peak contained most of the PCP displacing activity as measured by its ability to inhibit 3H-PCP. An aliquot of the most active material was hydrolyzed in acid and the amino acid composition was determined using OPA detection. It was determined that the peptide contained approximately 26 amino acids, in close agreement with the molecular weight predicted by Sephadex gel filtration studies. N-terminal analysis revealed that the peptide was blocked at this site. The nature of this blockade is yet to be determined. Studies are under way to determine the amino acid sequence of the peptide. [Pg.43]

Finally, radiochemical methods of analysis may be used to follow rates of detritiation. This method is particularly useful for very slow reactions (where it is impractical to collect data for any appreciable extent of reaction) as an initial rate approach may then be employed. Separation difficulties, at least for aqueous solutions, may be overcome by using the freeze-drying method or the more recent countercurrent dialysis and Sephadex gel filtration techniques. ... [Pg.4]

Sephadex Gel Filtration. The most common type used is the Sephadex G-200. The emergence soon after the void volume of a massive peak which is more prominent than the other peaks is compatible with a diagnosis of Waldenstrom macroglobulinemia. The test is a very useful aid in the diagnosis of Waldenstrom macroglobulinemia. [Pg.213]

There is some evidence that localized copper imbalance is related to rheumatoid arthritis (RA) conditions. Low molecular weight copper complex concentrations in plasma and synovial fluids are increased as part of the body s natural response to RA. Secondly, when such increases are induced by copper administration, they have a powerful antiinflammatory effect. More data are required to show the medical significance of these changes. Three parameters which should be reported are (i) total copper levels (by atomic absorption spectrophotometry) (ii) total ceruloplasmin levels (by autoimmune assay) and (iii) the relative molecular weight distribution (by Sephadex gel filtration and ultrafiltration). [Pg.759]

D-isomer. It has been possible to separate and identify the different variants of alkaline phosphatases in human placenta by Sephadex-gel filtration, sucrose density-gradient centrifugation, and ultracentrifugation, and in these studies starch-gel electrophoresis has proved to be unique in characterization of different isoenzyme fractions. Gel filtration chromatography has been widely used to separate high and low molecular mass phosphorus in sediments, soil solutions, and preconcentrated surface water. Steward and Tate (1971) showed, for example, that Sephadex G50 could be used to separate high molecular mass phosphorus that corresponded with the molecular weight of myo-inositol hexakisphosphate. Others used Sephadex gel filtration separations to demonstrate the presence and importance of inositol phosphates in preconcentrated lake water (Eisenreich and Armstrong, 1977), algal cultures (Minear, 1972),... [Pg.9]
