Semi-automated approaches to NOESY assignment [85-87] use the chemical shifts and a model or preliminary structure to provide the user with the list of possible assignments for each cross peak. The user decides interactively about the assignment and/or temporary removal of individual NOESY cross peaks, possibly taking into account supplementary information such as line shapes or secondary structure data, and performs a structure calculation with the resulting (usually incomplete) input. In practice, several cycles of NOESY assignment and structure calculation are required to obtain a high-quality structure. [Pg.53]

Qualitative methodology uses small samples of subjects and the method of (usually) personal interviews, based on structured or more usually semi-structured questionnaires. The interviews are tape- or video-recorded and then transcribed and analyzed for patterns and categories in students thinking. Data treatment can also be done by commercial packages for qualitative analysis. This methodology provides the possibility for in-depth monitoring and study of students ideas and understandings about scientific concepts. The use of LC-ARC or 96-well microtiter plates containing solid scintillant, in contrast, represents low-cost methodologies to improve the sensitivity of the semi-quantitative analysis of metabolites. The LC-ARC and 96-well microtiter plate approaches have similar LOD for 14C and 3H and both can be used in combination with MS to generate quantitative and structural data. Both approaches are particularly useful in cases where the sample is limited. The throughput of LC-ARC is higher than for the plate approach due to the need to dry and count the plates offline. Semi-empirical NDDO methods have been used for the study of various heterocycles <1995JMT51>. A belief that highly accurate values are not essential for valid conclusions to be drawn and the time advantage of semi-empirical over ab initio methods were factors governing the choice of these procedures.
