Sandoz additives

The chemical industry manufactures a large number of antioxidants (qv) as well as uv stabilizers and their mixtures with other additives used to facilitate resin processing. These companies include American Cyanamid, BASE, Ciba—Geigy, Eastman Chemical, Elf Atochem, Enichem, General Electric, Hoechst—Celanese, Sandoz, and Uniroyal, among others. The combined market for these products in the United States exceeded 900 million in 1994 and will reach 1 billion in the year 2000.

Hydroxymethyl-LSD, 1-dimethylaminomethyl-LSD and 2-iodo-LSD. These three additional compounds are shown here because they were described in a synthetic flurry that followed the discovery the activity of LSD. But at the moment I know neither their internal Sandoz codes nor if they had ever been explored in man. This is a kind of frustrating catch-all entry, in that the long index will send you here, and once here you realize that nothing is known. Well, at least the compounds are known, and perhaps there is something in the Sandoz vaults that might be interesting. I do not have access to them. 

Viscose or rayon A well-known inherently FR viscose fiber is Viscose FR, marketed by Lenzing. The fiber is produced by adding Sandoz 5060 (Clariant 5060)-bis(2-thio-5,5-dimethyl-l,3,2-dioxa-phosphorinyl)oxide in the spinning dope before extrusion. As this additive is phosphorus based, it is similar to other phosphorus-based FRs in terms of mode of action (condensed phase). 

Voclosporin (ISA-247, R1524) 88 (Isotechnika) is being evaluated in a Phase III trial for the treatment of psoriasis,250 as well as a Phase III trial by Lux Biosciences as for the treatment of uveitis (coded as LX211, Luveniq ).251,252 In addition, voclosporin 88 has completed a Phase lib trial for the prevention of kidney graft rejection. Voclosporin 88253,254 is a slightly more potent but less toxic semi-synthetic derivative of the fungal-derived immunosuppressant cyclosporin A 89, which has the same mechanism of calcineurin inhibition. Cyclosporin A 89 was first isolated from Tolypocladium inflatum by workers at Sandoz and its structure was published in 1976.255,256...

In a similar fashion, it has been possible to isolate different conformations of the immunosuppressive drug, cyclosporin A. By preparing anhydrous THF solutions of CsA, with and without 0.4 M LiCl, it was possible to restrict the peptide ring system conformation in CsA to trans and cis conformers. Addition of these conformers separately to the assay buffer enabled the dissociation constants of both the cis and trans amide bond conformers in CsA for inhibition of PPIase to be determined. The trans conformer (2337t) is very active the cis conformer (23.37c) is not, and these plus other data were used to show that CsA adopts a conformation in water that is very close to the enzyme-bound CsA conformation. Subsequently, Wenger and coworkers at Sandoz (now Novartis) showed by NMR experiments that a 3-substituted CsA derivative, e.g. (23.38) (Fig. 23.8) adopts a conformation in water that is essentially identical...
