Optimization of sample preparation depends upon the nature of the sample, the information required and the type of mass spectrometer available. It is desirable to minimize salts and detergents, and if buffers are unavoidable these should be volatile whenever possible, e.g. ammonium formate or ammonium bicarbonate.
