In a subacute inhalation toxicity study, rats of both sexes were exposed for 6 hours/day, 5 days/week for 4 weeks. Recovery groups were kept for an observation period of 14 days without treatment. At 891, 441, or 142 ppm hemolytic anemia was observed. Mild hemolytic anemia was found in female rats exposed to 100 ppm, but it had disappeared after the 14-day recovery period. [Pg.412]

The usual GLP 30- or 60-day repeat-dose toxicology study with a recovery group offers an opportunity to perform a more systematic investigation of the more subtle pharmacodynamic or toxicologic effects of biopharmaceuticals than those endpoints usually incorporated into such protocols. Some of these demand tissue samples, but many involve noninvasive biomarkers that can be carried forward into early phase human studies. These might include CNS assessments, inflammation and immune activation or suppression, cell proliferation or apoptosis in tissue samples, and end-organ toxicities. [Pg.321]

Recovery group included in the study and data generated are sufficient to determine the degree of reversibility. [Pg.79]

Potential carcinogenicity of PEGS has been examined in a dietary 2 year bioassay with rats. In male and female rats exposed to 20 mg kg there was a significant increase in the incidence of hepatocellular adenomas while in females there was also an increase in hepatocellular carcinomas. In addition, there was a significant increase in thyroid follicular cell adenomas and carcinomas in males in a recovery group treatment group. [Pg.2513]

Notes In a comparison of the clinical characteristics at baseline in the patients with iodine restriction, the pretreatment iodine intake in the recovery group (patients who recovered from hypothyroidism to the euthyroid state spontaneously after 12-week iodine restriction) tended to be higher than that of the nonrecovery group (patients who did not recover from hypothyroidism to the euthyroid state after 12-week iodine restriction), but the difference was not statistically significant. ... [Pg.761]

The test and control diets were presented to their respective groups on day 0 of the study. All the low- and mid-dose animals and half of the control and high-dose animals (non-recovery groups 1-8) were exposed to their test or control diet for at least 28 days, prior to necropsy on day 31. The remaining control and high-dose animals (groups 9-12) constituted the recovery group and were exposed to... [Pg.292]

Under the conditions of this study and based on the toxicoiogicai end-points evaluated, the NOEL was the highest dose tested—nameiy, 1100 mg/kg, equivaient to 92 and 97 mg/kg bw per day in the non-recovery group male and female rats, respectively, and 94 and 97 mg/kg bw per day in the recovery group male and female rats, respectively (Everds, 2005 Funk, 2005 Merkel, 2005). [Pg.294]

Figure 6 Effects of chronic hypoxia (CH) on type I cell [Ca ]i responses to acute hypoxia challenge at 3, 11, and 18 days from CH ( ) and control (O) rats. In panel c, A represents data from a recovery group reared in CH from hirth to 11 days and then in room air from 11 to 18 days. CH from hirth abolished postnatal development of [Ca ] responses to h.3fpoxia. After 11 days in h3fpoxia, returning to room air for 1 week resulted in partial recovery of [Ca ]i response to h.3fpoxia (c). indicates significant difference from CH p < 0.05).
