Observations are made in metaphase cells arrested with a spindle inhibitor such as colchicine or colcemid to accumulate cells in a metaphase-like stage of mitosis (c-metaphase) before hypotonic treatment to enlarge cells and fixation with alcohol-acetic acid solution. In a double-bUnded study, both the Investigator and the subjects are unaware of whether they receive the drug or the placebo. The randomization code is held in confidence and is opened at the end of the trial for data analysis or in cases where adverse events occurred.

C) The achievable rates computed for SCS can be obtained only for codewords of infinite length. In practice, the codeword length is finite. With help of Gallager s random-coding exponent, we analyze the influence of codeword length on the performance of SCS in Sec. 5. 

Blindness can also be destroyed by accidental breaking of the randomization code. With good design and adequate safeguards, however, this should not occur. 

However, the method of labeling the study drugs should allow for this to be done without revealing the randomization code for the entire study. One such technique is for each medication container to have a two-part label, one of which is a sealed, tear-off section containing the identity of the drug. At the time the patient begins treatment, the tear-off section is detached and affixed to the patient s case report form and can be opened in an emergency. 

Administrative interim analysis should provide feedback on whether the study is being executed as designed and whether the data are being thoroughly and accurately collected. This is a gauge of the conduct of the study, not of the results. Randomization codes need not be broken. 

Storage of randomization codes and code-breaks, and access control... 

Randomization codes are used for packaging and labeling study medications. The systems that are used to generate labels for the treatment bottles may be independent of those that produce the randomization. 

The randomization codes may now be entered allowing the statistician to review the data in an unblinded fashion. Any pharmacokinetic data are also loaded at this time. Data management then freezes the database. No changes may be made to the existing database and no new data may be added. 

In the simplest case of parallel-group study, a group of patients presenting sequentially are randomized to one of two equally sized treatment groups, until a prospectively determined total number of patients has been recruited. All these patients are followed for a predetermined period of time, or until some end point is achieved. The database is quality assured and locked before the randomization code is broken. The patients are then sorted according to their treatment, the end point measurements are subjected to a statistical test and an interpretation of the effect (or absence thereof) of the drug is made. What could possibly go wrong ...

The project statistician will then merge the file of the randomization codes to the database to generate the analysis datasets. 

Keep the randomization codes and opening them, if necessary... 

These are the unintended reactions of the subjects as a consequence of taking the drug or placebo. Subjects are checked and, if the adverse events are serious, subjects may be temporarily removed from the trial. If there is a persistence of adverse event, the subject may be withdrawn from the trial. The randomization code may be broken (opened) to determine whether the subject has been given the drug or placebo. 

Procedures should be established for the generation, distribution, handling and retention of any randomization code used in packaging investigational products. 

A system should be implemented to allow for a proper identification of the blinded products. The system, together with the randomization code and randomization list must allow proper identification of the product, including any necessary traceability to the codes and batch number of the product before the blinding operation.
