Another basic issue to be considered was represented by the influence of PHB on the biodegradability of PVA in the presence of specific PVA-degrading microorganims, by considering that biodegradation of the polyester and of the polyhydroxylated polymer is strictly mediated by PHB-depolymerase and PVA-oxidase specific enzymes, respectively. In this connection, the biodegradability of both a graft copolymer and a cast blend was ascertained in the presence of a select bacterial culture able to utilize PVA. [Pg.338]

A flow injection optical fibre biosensor for choline was also developed55. Choline oxidase (ChOX) was immobilized by physical entrapment in a photo-cross-linkable poly(vinyl alcohol) polymer (PVA-SbQ) after adsorption on weak anion-exchanger beads (DEAE-Sepharose). In this way, the sensing layer was directly created at the surface of the working glassy carbon electrode. The optimization of the reaction conditions and of the physicochemical parameters influencing the FIA biosensor response allows the measurement of choline concentration with a detection limit of 10 pmol. The DEAE-based system also exhibited a good operational stability since 160 repeated measurements of 3 nmol of choline could be performed with a variation coefficient of 4.5%. [Pg.171]

Acetylcholineesterase and choline oxidase Prepared by mounting a carbon fiber (200 pm diam) in a glass capillary with silver paste and epoxy resin, electrochemically pretreating the electrode from 0 to 1.2 V for 15 min, and dipping the electrode in 11% PVA-Styryl pyridinium solution containing AChE and ChO. The calibration graph was rectilinear from 0.2 to 1 mM of ACh. The response time was 0.8 min. [76]... [Pg.34]

Acetylcholineesterase and choline oxidase Enzyme membrane in H20 was treated with 11% solution of PVA-SbQ (polyvinyl alcohol) with styryl pyridinium groups. Mixture was spread on a cellulose nitrate membrane and air dried. The membrane was exposed to UV radiation for 3 h and stored at 4°C. The enzyme membrane was fixed with a Pt electrode. Sample was dissolved in phosphate buffer and measured. The best results were obtained at pH 8 and at 30°. The calibration graph was rectilinear for 5mM ACh. The storage stability of the dry membrane was excellent. [77]... [Pg.34]

In the case of PVA, which is one of the few carbon-chain polymers that does not need to be abiotically peroxidised before biological attack, polyketones are the initial products and these are catabolysed to carboxylic acids and bioassimilated by bacteria e.g. Pseudomonas). Kawai has shown (personal communication) that the PEGs behave rather similarly and are bioassimilated by Sphingomonas. The active enzymes are believed to be PEG dehydrogenase coupled with cytochrome c, the oxidase enzyme of the baeterial respiratory system. Since the polyethers are also very peroxidisable abiotically, abiotic peroxidation may also play a part in the overall process. [Pg.101]
