Proinflammatory cytokines source

In the very early phases of the acute inflammatory response most of the cells invading the damaged area are polymorphonuclear neutrophils, also denoted as PMNs, which serve as initial line of defense and source of proinflammatory cytokines. These cells, which usually live for 4-5 days, circulate in the blood until they are attracted by chemokines into injured tissues. Whereas physical injury does not recruit many neutrophils, infections with bacteria or fungi elicit a striking neutrophil response. The characteristic pus of a bacterial abscess is composed mainly of apoptotic (apoptosis) and necrotic PMNs. Emigration of neutrophils from the blood starts with a process denoted as margination where neutrophils come to lie at the periphery of flowing blood cells and adhere to endothelial cells (Fig. 1). L-Selectin is expressed... [Pg.628]

In endotoxinemic mice IL-10 treatment inhibits proinflammatory cytokine release and leads to a reduction in LPS toxicity (G8, H28). IL-10 does not influence the LPS-induced production of IL-6 in vivo (M9). This suggests that IL-10 could differentially regulate TNF and IL-6 production by macrophages in vivo, in contrast to data obtained in vitro (Wl), or that cell types other than macrophages are a major source of IL-6 in vivo and are resistant to IL-10 (S23). [Pg.66]

In 2004 Okrm and colleagues studied the role of cytokines and growth hormone in the regulation of sleep and narcolepsy. Collectively, proinflammatory cytokines and human growth hormone (HGH), were found to be possibly involved in the development of or the exacerbation of narcolepsy (a disabling neurological sleep disorder characterized by excessive daytime sleepiness and abnormal sleep manifestations). As the role of the immune system in sleep becomes clearer, a better interpretation will be available to assess why narcoleptics have altered cytokine and HGH levels. Future studies should be conducted to fuUy characterize the sources, patterns, and significance of alterations in patterns of proinflammatory cytokines and HGH in narcolepsy. ... [Pg.652]

The mature peptide is derived from a pre-pro-form, which consists of 170 amino acids, being located between amino acid positions 125 to 152 within the prepro-form. Antigenic stimulation results in the early production of proinflammatory cytokines by activated macrophages, followed by the secretion of anti-inflammatory factors, including Aviptadil, from sources such as peptidergic innervations or the immune cells themselves. Aviptadil inhibits the production of the proinflammatory factors such as tumor necrosis factor alpha (TNF-a), interleukin 6 (IL-6), IL-12 and nitric ox-... [Pg.1744]

Maternal infection. Mild infection transmitted to the fetus is another source of possible subclinical changes in brain development.24 Clinical and epidemiological studies have shown associations between maternal infection, fetal brain damage, and increased levels of proinflammatory cytokines, a class of regulatory proteins secreted especially by cells of... [Pg.100]
