Pooled sampling

Pome Fruit Types. As with citrus fruit types, the method of sample preparation was modified for the parathion studies. In the earlier studies the DDT-treated apples and pears were scrubbed in a warm 10% solution of trisodium phosphate, and all the peel was removed from the water-rinsed fruit with a household-type potato peeler. The pooled samples of peel and pulp were then processed independently to recover the contained toxicant for subsequent estimation. 

Mean concentration of THg in tissue pooled samples (n = 34) was 1.49 0.22 (ig/g dw, ranging from 0.056 to 3.11 pg/g. The highest level was found at the factory site, located close to the waste dumping site. This result indicates an important mercury uptake by the specimens located at the hot spot and a limited redistribution to other areas close to the opposite river bank.

It was reported that the distribution and activities of esterases that catalyze pyrethroid metabolism using several human and rat tissues, including small intestine, liver, and serum, were examined [30]. The major esterase in human intestine was hCE2. //c/n.v-Permethrin was effectively hydrolyzed by pooled human intestinal microsomes (five individuals), while deltamethrin and bioresmethrin were not. This result correlated well with the substrate specificity of recombinant hCE2. In contrast, pooled rat intestinal microsomes (five animals) hydrolyzed trans-permethrin 4.5 times slower than the human intestinal microsomes. Furthermore, pooled samples of cytosol from human or rat liver were ca. half as hydrolytically active as the corresponding microsome fraction toward pyrethroids however, the cytosolic fractions had significant amounts (ca. 40%) of the total hydrolytic activity. Moreover, a sixfold interindividual variation in hCEl protein expression in human hepatic cytosols was observed. 

Rapid, accurate SNP validation can be carried out using a sample-pooling technique—allele frequency—that rapidly screens and confirms the presence of an SNP and its allelic frequency in patient populations. Conventional technologies typically analyze each SNP in each individual of the population in question, and individual results are then consolidated to yield the overall SNP allele-frequency distribution. MS-based technology is able to determine SNP allele frequencies with high precision in pooled samples, thereby replacing hundreds of individual measurements with one consolidated analysis. To that end, DNA is isolated, purified, and quantitated. A pool of DNA is formed from a high number of different individuals, amplified, and mass measured.

With the advent of API sources, LC/MS/MS allows the facile development of quantitative methods that are sensitive, selective, robust, and amenable to the rapid analysis of a majority of small molecules. In order to achieve high-throughput bioanalysis in support of pharmacokinetic studies, many approaches have been reported utilizing automated sample preparation and reducing analysis time by pooling samples, parallel analysis, and fast chromatography. 25,26,152,153...

Di(2-ethylhexyl) phthalate was found (mg/kg wet weight) in the following commercial fish (pooled samples of 10 individuals each) herring (fillets), 4.71 mackerel (fillets), 6.50 cod (liver), 5.19 plaice (fillets), < 0.010 and redfish (fillets), < 0.010 (Musial et al., 1981).

Using aseptic technique, open the required number of vials, vor-texing each prior to removing an aliquot. Pool the samples aseptically in a sterile, pyrogen-free vial. Eor in-process sample, treat the bulk solution as a pooled sample. 

A pool of plasma is composed by mixing residual amounts of plasma left over from analyses, thereby ensuring the anonymity of the pool sample. Each series of analyses should contain at least one pool plasma sample. The results of 8-10 selected amino acids of the pool are plotted in a Shewhart plot to make deviations from the previously established target values visible. Actions following these deviations will have to be devised by the laboratory staff according to standard quality control rules.

Of compounds tested in the rat, only phenylethylamine produced a prolonged rise in blood pressure. Mixed picrates of blood extracts were a preparation of hypertensive blood extracts purified by the formation of picrates. These gave an immediate pressor response. Pooled pressor blood extracts indicate extracts taken from a number of samples of hypertensive arterial blood and concentrated. Pooled samples from normal subjects gave no such response.
