Partial and Total Synthesis

The isolation of the active metabolites of vitamin D from biological tissues is extremely laborious and affords only small quantities of material. Development of synthetic approaches to these materials was thus necessary to provide them and related analogs in adequate quantity for elucidation of their mode of action. The new chemistry required was that in which new functionalities, usually hydroxyl groups, could be [Pg.74]

The emphasis in work related to diterpenoid alkaloids has shifted from structure determination to partial and total synthesis. X-Ray crystallography played the major role in the structure work and in interpretation of unusual chemistry. However, some interesting new chemical and spectroscopic observations, and analyses of mass spectra, have been reported. [Pg.343]

The challenge of effecting in the laboratory the rearrangement from an atisine to a lycoctonine skeleton is very relevant for both partial and total synthesis. Attempts to do this by solvolysis of derivatives with either configuration at C-15 of compounds derived from atisine had given products with the wrong skeleton (e.g. 133 134). However, Johnston and Overton have found that pyrolysis... [Pg.368]

Topics covered in this b3ok include the partial and total synthesis, biosynthesis, and structure-activity relationships of the principle classes of /3-lactam antibiotics, including penicillins, cepfialosporins, clavulanic acids and olivanic acids, and their interactions with bacteria. [Pg.467]

The chemistry of natural products encompasses their isolation, structure elucidation, partial and total synthesis, elucidation of their biogenesis, and the biomi-metic synthesis of N. p. Major breakthroughs in analysis were, e.g., the structural clarifications of morphine, lignin, insulin, estrones, and cholesterol as well as the elucidation of the biosyntheses of terpenoids, morphine, penicillin, chlorophyll, and vitamin B 2. Major advances in synthetic chemistry were, e.g., the total syntheses of camphor, hemin, quinine, saccharose, tropine, stryehnine, chlorophyll, vitamin B 2, erythromycin, taxol and palytoxin. Numerous N. p. of the so-ealled ehiral pool are used as starting materials for the synthesis of optically active compounds or serve (in the form of their derivatives) as catalysts for enantioselective syntheses. [Pg.424]

Since all the most important natural steroids have already been obtained by total synthesis, each reaction in the steroid series can in fact now be regarded as a "formal total synthesis" of the products formed in it. Such a situation requires the drawing of the clearest possible boundaries between the partial and total synthesis of steroids. In our opinion, the term total synthesis can be applied only to those investigations in which the construction of the side chains distinguishing the classes of steroid compounds was connected organically with the construction of the steroid skeleton (as is the case in the syntheses of aldosterone and conessine). From this point of view, it must be regarded as incorrect to describe as total syntheses, for example, the syntheses of equilin, diosgenin, and tomatidine these syntheses deliberately start from natural steroids and must be classified as partial syntheses. In view of what has been said, in this book our main attention is devoted to the construction of the steroid skeleton and not to the introduction of side chains. The broad development of the chemistry of the heterocyclic steroids has made it necessary also to consider the main methods for the total synthesis of azasteroids, oxasteroids, and thiasteroids. [Pg.372]

Production. (—)-Citronellol is still obtained mainly from geranium oil by saponification followed by fractional distillation. Although of high odor quality, it does not possess the true (—)-citronellol odor due to impurities. Much larger quantities of (+)- and (zb)-citronellol are used and are prepared by partial or total synthesis. [Pg.32]

Production. Many industrial processes exist for the production of menthols. For (—)-menthol, isolation from peppermint oil (see Mint Oils) competes with partial and total syntheses. When an optically active compound is used as a starting material, optical activity must be retained throughout the synthesis, which generally consists of several steps. Total syntheses or syntheses starting from optically inactive materials require either resolution of racemic mixtures or asymmetric synthesis of an intermediate. Recently used processes are the following ... [Pg.53]

Synthetic work in the steroid field remains an active area of research. A number of useful approaches to the steroid system, by either partial or total synthesis, have been reported during the year under review. The purpose of the present chapter is to put emphasis on original synthetic processes and sequences of general applicability. No mention will be made of particular reactions, the application of physical methods, biosynthetic studies, or microbiological reactions. [Pg.329]

Conclusions - Efforts toward the synthesis of the anthracycllnes have provided a chemical harvest which Is already bounteous and continues to be reaped. Overall yields for the total synthesis of the Intact, chiral natural products la-3a, as well as and their analogs, are now on the order of 5% In the best cases. Whether total (or partial) synthesis will emerge as the practical solution for the future Is, however, a question whose ultimate answer depends on many as yet Incompletely resolved factors which include the relative economics of fermentative and total synthesis, the potential therapeutic superiority of non-natural anthracycllnes, and future advances in total synthesis. It Is the opinion of the author that at the present time fermentation remains the preparative method of choice, but this verdict Is much less certain than It would have been five years ago. Five years hence such a judgment may be Indefensible. Synthetic chemists would not be distressed. [Pg.297]

All these results underlined the remarkable, nearly perfect fit of the natural product into a naturally unrelated biological target, and directed most of the synthetic efforts towards partial or total synthesis of paclitaxel rather than of its structural congeners. In the following three sections, the specific achievements of the three reactions catalyzed by organometallic complexes are presented, showing the key transformations that provide the key building blocks for paclitaxel. [Pg.181]

As is true of other fields of science, natural products chemistry is in a state of transition and is undergoing a profound change in character. Being intimately related to nature and natural phenomena, plants, animals, food, poisons, diseases, etc., the chemical studies in the natural products area can be traced back thousands of years. Traditional natural products chemistry necessarily started with the isolation and purification of a compound, followed by structure determination, and culminated in partial or total synthesis. [Pg.14]

Partial or total synthesis of mentioned compounds are presented according to their chronological order of publication and, for a same year by alphabetical order of the first author. [Pg.136]
