Nucleotides ordering

DNA sequencing means a reading of the nucleotide order of a piece of DNA. The main purpose of the Human Genome Project (HGP) was to determine the nucleotide composition of the human genome. This was accomplished with an immense international effort for 3 billion between 1990 and 2003 [30]. It also became apparent how little we knew about the human genome and how valuable it would be if a human genome had been sequenced for a much cheaper price and less time. DNA from a gene contains hundreds to thousands of nucleotide units for which the sequence is needed in order to interpret its code. Sequencing methods require only small amounts (5 (tg) of purified DNA, which can be produced by cloning. Automated sequencers are available that can daily sequence DNA containing hundreds of nucleotide units. [Pg.329]

In the cytoplasm, the mRNA attaches to a ribosome and acts as a template for the construction of a protein with the proper amino acid sequence (a process known as translation ). Single amino acids are brought to the ribosome by transfer RNA molecules (tRNA) and added to the growing amino acid chain in the order instructed by the mRNA. Each time a nucleotide is added to the growing RNA strand, one molecule of ATP is broken down to ADP. Each time a tRNA binds an amino acid and each time the amino acid is added to the protein, additional ATP is broken down to ADP. By structural complementarity, dicationic l,4-diazabicyclo[2.2.2]octane (VII) provides an appropriate recognition site for phosphate ions and two stearyl side chains attached to the amines add lipophilic properties 59,60). Such a carrier model can selectively extract nucleotides from aqueous solution to chloroform solution via lipophilic salt formation. The order of nucleotide affinity is ATP > ADP > AMP.
