SELECTED EXAMPLES OF DRUG ACTION AT SOME COMMON TARGET AREAS [Pg.156]

I.2.4.3 Nucleic Acid Synthesis Inhibitors This mechanism is best exemplified by quinolone antibiotics. The basic structure of aU quinolone antibiotics is similar, with an exocyhc oxygen at position 4. The core structure is ... [Pg.52]

The intercalating drugs and nucleic acid synthesis inhibitors discussed in the preceding sections interfere indirectly with every phase of nucleic acid action because the DNA is rendered nonfunctional. Other drugs, discussed below, affect the regulation of protein synthesis even when the genome, the DNA structure, is intact. Such activity can be the result of interference either with transcription of messenger RNA or with translation of the mRNA to protein. [Pg.453]

It is known that protein synthesis is necessary for BR-induced effects in root tissue (25), and BR-treatment increases nucleic acid and protein synthesis in bean stem (26). In the pea stem segment, kinetic studies with selected protein and nucleic acid synthesis inhibitors showed no evidence for competitive inhibition in polypeptide chain elongation, post-transcriptional polyA addition to heterogeneous RNA, DNA-dependent RNA synthesis, or of the DNA-dependent RNA polymerase... [Pg.258]

Bacterial infections are treated with antibiotics. There are many antibiotics available, but they fall into three major groups based on their mode of action inhibitors of bacterial nucleic acid synthesis inhibitors of cell wall synthesis and inhibition of bacterial protein synthesis. Resistance of bacteria to commonly-used antibiotics has become a major problem necessitating the development of new antibiotics. Tuberculosis infection is difficult to treat and requires a combination of at least three different antibiotics. [Pg.173]

The experimental approach in most of the systems studied so far was to follow the greening process at different levels (ultrastructure, composition of membranes, development of activity) and try to establish a correlation between the different parameters observed which will lead to the formulation of explanatory hypotheses. The validity of the hypotheses can then be assessed by inducing alteration in the process at one level and studying the effect at the other levels. The use of protein and nucleic acid synthesis inhibitors has so far been the method of choice. Cycloheximide was extensively used to block protein synthesis by cytoplasmic 80 S type ribosomes, while chloramphenicol, spectinomycin, and lyncomycin were used to inhibit protein synthesis by the chloroplast ribosomes. Actinomycin D and, more recently, rifampicin have been used as inhibitors of RNA synthesis. Initially, rifampicin was considered to be specific for the DNA-dependent RNA polymerase of the chloroplast. Although it appears that in some algae, such as Chlamydomonas and Acetabularia, rifampicin has a reasonable degree of specificity, its action against RNA polymerase of the chloroplast partially purified from plants was not found to be specific. ... [Pg.287]
