The top section indicates the Old (and in some cases, current) way in which pharmaceutical companies take Hits to lead development candidate through discovery and early preclinical development testing. Essentially, no high-throughput formal lead optimization exists in this model but is developed for backup compounds by using biomarkers identified during lead compound failure in vivo. The bottom section indicates the New way forward (used now by most major pharmaceutical companies). In this model, a formal in vitro PLOT battery is inserted early in development before preliminary in vivo ADME-Tox screening and subsequent lead development candidate selection.

Together with the advent of the protease inhibitors. Saquinavir, Indinavir, and Ritonavir, the recent approval of Epivir for use in combination with Retrovir marks a new way forward in AIDS therapy. 

Implementation of the Kohn-Sham-LCAO procedure is quite simple we replace the standard exchange term in the HF-LCAO expression by an appropriate Vxc that will depend on the local electron density and perhaps also its gradient. A number of approaches to predict ionization based on structure have been published (for a review, see [53]) and some of these are commercially available. Predictions tend to be good for structures with already known and measured functional groups. However, predictions can be poor for new innovative structures. Nevertheless, pfCa predictions can still be used to drive a project in the desired direction and the rank order of the compounds is often correct. More recently training algorithms have also become available which use in-house data to improve the predictions. This is obviously the way forward. 

It appears to us that the way forward to further industrial application of solid base catalysts may emerge from the discovery of unusual selectivity effects or new reactions that require well-balanced base and acid-base pair sites with relatively inexpensive solid catalysts that last long enough to process large quantities of reactants per unit mass of catalyst. 

Behavioural change is not always the appropriate way forward. If you feel the new behaviour will be too alien for you to adapt to, some prior work on your psychology may be indicated. 

Vortices, Explosive Powered. During WWII in Ger, a Dr Zippermeyer, working at the Speer Ministry s Research Establishment near Lofer, attempted to duplicate in miniature the effects of tornadoes using expl powered vortices. Although man-made vortices had heretofore been produced using compressed air, Zippermeyer proposed to power the vortex in an entirely new way. He proposed to shoot powdered coal in a projectile. In the center of the mass of powdered coal, he placed a charge of low expl, so that, upon initiation, the coal dust would have a forward component of velocity due to the motion of translation of the projectile and a lateral...

There are many excellent books on Excel in general, and the package in itself is associated with an extensive system. It is not the purpose of this text to duplicate these books, which in themselves are regularly updated as new versions of Excel become available, but primarily to indicate features that the user of advanced data analysis might find useful. The examples in this book are illustrated using Office 2000, but most features are applicable to Office 97, and are also likely to be upwards compatible. It is assumed that the reader already has some experience in Excel, and the aim of this section is to indicate some features that will be useful to the scientific user of Excel, especially the chemometrician. This section should be regarded primarily as one of tips and hints, and the best way forward is by practice. There are comprehensive help facilities and Websites are also available for the specialist user of Excel. The specific chemometric Add-ins available to accompany this text are also described.
