The very high activity found in the molecular layers cannot be interpreted at present, but it may be noted that these layers contain local neurones morphologically somewhat similar to the basket cells (Ramon y Cajal, 1893 Lorente de No, 1934), but with unknown function.

Figure 5.8 shows some typical neuron morphologies. The motor neuron is found in the spinal cord. It is an efferent type bringing information from the central nervous system (CNS) to a muscle or gland, for example. The receptor type is an afferent neuron with a special direct axon coupling between the receptor and the CNS. The pyramid morphology is found in the cortex of the brain.

After 9 days in culture, wash cells twice with phosphate-buffered sahne (PBS, pH 7.4) to remove media and cell detritus. This immunofluorescence protocol is similar to many protocols used extensively in the scientific community, and variations would likely not affect the ability to analyze changes in neuronal morphology.

Alternatively, dissociated cells can be collected by centrifugation (2500 rpm for 2 minutes), resuspended in medium containing 2 pg/ml cytochalasin B, and plated on glass cov-erslips. The cytochalasin acts to inhibit cell division and promote the growth of giant neurons the drug can be removed after the second day of culture or left on with no ill effects (Wu et al. 1990 Saito and Wu 1991). The cultures are maintained in humidified chambers at room temperature (20-23 >C). Under these conditions, the majority of cells (>80%) differentiate neuronal morphology.
