Neomycin resistance

Within the first days under selecting conditions you should observe cell death or an appearance change in a part of the colonies. This serves as an indicator that G418 treatment is deleting the cells which are not expressing the neomycin resistance gene. [Pg.321]

Retrovirus vectors were subjected to the first clinical trial on human gene therapy to correct adenosine deaminase (ADA) deficiency (32). White-blood cells isolated from patients were infected ex vivo with an MLV-based vector expressing ADA and a neomycin marker gene. After selection with G418, neomycin-resistant cells were isolated and reintroduced into patients. The treatment improved the physical condition of the patients and the ADA-containing provirus was stable in the blood for several years. [Pg.339]

Gene transfer of mouse and human CD34+ HSCs, which were genetically modified by a retroviral virus encoding ADA cDNA and the neomycin-resistance marker gene, have been extensively characterized in mouse models without reports of toxicity. Furthermore, none of the preclinical studies for ADA-SCID have demonstrated evidence of cancer in animals treated with this same gene transfer approach [515137], [666652], [666655]. [Pg.81]

R factor mediated kanamycin resistance in strains of Ps. aeruginosa has been observed by a number of workers. The pseudomonas R factor RPl (142) carries the neomycin-kanamycin phosphorylating enzyme [205]. A study of the prevalence of R factor carrying strains of Ps. aeruginosa in Japanese hospitals showed a number of strains capable of transferring both kanamycin and neomycin resistance [206]. [Pg.375]
