Membrane synthesis monitoring

Confocal Raman spectroscopy is also a powerful tool for the monitoring of membrane synthesis. It has been used for instance to demonstrate in situ the... [Pg.146]

In addition to the insoluble polymers described above, soluble polymers, such as non-cross-linked PS and PEG have proven useful for synthetic applications. However, since synthesis on soluble supports is more difficult to automate, these polymers are not used as extensively as insoluble beads. Soluble polymers offer most of the advantages of both homogeneous-phase chemistry (lack of diffusion phenomena and easy monitoring) and solid-phase techniques (use of excess reagents and ease of isolation and purification of products). Separation of the functionalized matrix is achieved by either precipitation (solvent or heat), membrane filtration, or size-exclusion chromatography [98,99]. [Pg.87]

The cytosol is the fluid compartment of the cell and contains the enzymes responsible for cellular metabolism together with free ribosomes concerned with local protein synthesis. In addition to these structures which are common to all cell types, the neuron also contains specific organelles which are unique to the nervous system. For example, the neuronal skeleton is responsible for monitoring the shape of the neuron. This is composed of several fibrous proteins that strengthen the axonal process and provide a structure for the location of specific membrane proteins. The axonal cytoskeleton has been divided into the internal cytoskeleton, which consists of microtubules linked to filaments along the length of the axon, which provides a track for the movement of vesicular material by fast axonal transport, and the cortical cytoskeleton. [Pg.10]

Reliance on enzyme activity as the monitoring device also limits study to enzyme-type proteins, a most unfortunate situation since many interesting proteins do not have catalytically assayable functions. Some proteins are regulatory elements which modulate synthesis or activity of enzymatically active proteins others are structural elements or proteins that can perform their function only when integrated into a membrane, that is, permeases. The functions and modes of operation for many of these proteins are being elucidated with the help of immunochemical assay procedures. [Pg.285]

Heterodisulfide (CoM-S-S-HTP) reduction - coupled to primary translocation. The reaction in which ATP is synthesized during methanol reduction to CH4 could be identified with energetically competent membrane vesicles of the methanogenic strain G61. These vesicles, which are orientated more than 90% inside-out, catalyzed CH4 formation from CH3-S-C0M by reduction with H2 (Reactions 7,8) and coupled this process with the synthesis of ATP [112]. CH3-S-C0M reduction generated a ApH (inside acidic) as monitored by acridine dye quenching protonophores and ATP synthase inhibitors exerted their effects in accordance with a chemiosmotic type of ATP synthesis [113],... [Pg.128]

In two variations of this basic procedure, transport of VSV G protein is monitored by different techniques. Studies using both of these modern trafficking assays and Palade s early experiments all came to the same conclusion in mammalian cells vesicle-mediated transport of a protein molecule from its site of synthesis on the rough ER to its arrival at the plasma membrane takes from 30 to 60 minutes. [Pg.704]

Daptomycin is a fermentation product having a cyclic lipopeptide structure. It is primarily active against Gram-positive infections, especially those involved in skin/skin structure infections. It is given IV but must be administered over a period of 30 minutes or more. It binds to cell membranes and causes depolarization, which interrupts protein, DNA, and RNA synthesis. Daptomycin is bactericidal. Although resistance can be achieved in vitro, resistance has been slow to emerge in the clinic. Patients should be monitored for muscle pain or weakness, because some incidence of elevated serum creatinine phosphokinase is associated with its use. A small number of clinical trial patients also developed conditions related to decreases in nerve conduction (e.g., paresthesias and Bell s palsy). Daptomycin is eliminated primarily by the kidney, so dose adjustment may be necessary in cases of renal insufficiency. [Pg.1647]

Real-time monitoring of enzymatic reactions is also useful for fundamental studies in synthetic biology. Bujara et al. [56] implemented ESI-QqQ-MS in the monitoring of a multi-enzyme reaction chain (Figure 13.7). This method may enable optimization of complex metabolic reaction networks for chemical synthesis. The on-line sampling system encompassed sampling of the reaction mixture via membrane, reduction of flow, dilution, and ionization. [Pg.326]

According to Mitchell s chemiosmotic theory [52,53] active transport, oxidative phosphorylation and ATP synthesis are driven by a protonmotive force which is made up of two components an electrical potential gradient and a pH gradient across the cell membrane. Agents which conduct ions across the membrane destroy the pH gradient and thereby reduce the protonmotive force. It is possible to measure the pH gradient and to monitor its attenuation by... [Pg.92]

Free amino functions on the spots can be stained with bromophenol blue (26) prior to the coupling reactions. This allows visual monitoring of the performance of all the synthesis steps. Thus, a standard membrane for SPOT-... [Pg.308]

Other applications in micro and nanoelectronics are been developed with the help of CNTs. One example is the development of sensors that are placed in situ in concrete for the evaluation of internal porosity [13], through the use of oriented nanotube membranes that vibrate when they are in the middle. For certain applications, the solution to this approach is the synthesis of films of aligned CNTs [14-17], enabling the manufacture of monitors and microwave generators. This is possible because of their ability to emit electron by field effect [18]. The applications of these films also include the development of polymeric solar cells the semiconducting properties of CNTs films with anisotropic morphology result in a route for the separation of pairs and conduction electrons/holes generated by photons [19]. [Pg.44]
