Meiotic mutation

Protocol III.C.3 is the method of choice when the number of female flies is limiting, as it often is when working with special stocks such as those carrying meiotic mutations. One can easily obtain enough egg chambers for a hybridization experiment from 6-12 females by this dissection method, and the different stages are more evenly represented, but it is more laborious. 

Taking advantage of the homothallic life cycle of yeast (Herskowitz, 1988), it is possible to generate and screen directly for recessive (and dominant) meiotic mutations. The basic scheme is as follows (Fig. 1) an appropriately marked homothallic diploid is grown to saturation in YEPD [2% (w/v) dextrose, 2%... 

In male rats 2,3,7,8-tetrachlorodibenzo-p-dioxin induced hyperplastic changes and sperm granulomas in the epididymis, but no apparent lethal mutations were noted during post-meiotic phases of spermatogenesis.

Carrier assignment, however, is not perfected. Linkage studies are complex, require participation of multiple family members, and are confounded by inaccuracy caused by meiotic recombination or germline mosaicism. Serum CK is, by definition, above the reference interval (defined as the 95th percentile of a healthy reference group of age-matched women) in 1 of 20 noncarrier women. Moreover, serum CK decreases in DMD carrier women as they age. Thus for DMD and/or BMD. families, identification of the mutation in the family best enables rapid and accurate direct mutation analysis for carrier and prenatal testing in that family.
