Mapping of eukaryotic chromosomes has involved additional methods which are discussed in Sections E and G,l. These incluse radiation hybrid mapping,104 use of meiotic recombination, identification of restriction fragment length polymorphisms (RFLPs described in Section E,7), and use of expressed sequence tags (ESTs), short DNA sequences deduced from mRNA molecules transcribed from the DNA.105 106... 

Mismatch repair was an essential component of the Holliday model for meiotic recombination in fungi (Holliday, 1965a). This states that recombination depends on the formation of relatively short stretches of hybrid DNA between homologous chromatids within which, if they spanned a region of heterozygosity, a mispaired base-pair would appear. Mismatch repair would determine whether or not aberrant segregations would be found in the tetrads formed on completion of meiosis.

Carrier assignment, however, is not perfected. Linkage studies are complex, require participation of multiple family members, and are confounded by inaccuracy caused by meiotic recombination or germline mosaicism. Serum CK is, by definition, above the reference interval (defined as the 95th percentile of a healthy reference group of age-matched women) in 1 of 20 noncarrier women. Moreover, serum CK decreases in DMD carrier women as they age. Thus for DMD and/or BMD. families, identification of the mutation in the family best enables rapid and accurate direct mutation analysis for carrier and prenatal testing in that family.

Meiotic recombination in diploid organisms requires extensive sequence homology between the recombining partners and is called homologous recombination. This term also describes certain recombinational events between bacterial chromosomes. Most bacterial homologous recombination processes share a common requirement for the RecA protein or its counterpart.
