In the 40 years since Olah s original publications, an impressive body of work has appeared studying carbocations under what are frequently termed stable ion conditions. Problems such as local overheating and polymerization that were encountered in some of the initial studies were eliminated by improvements introduced by Ahlberg and Ek and Saunders et al. In addition to the solution-phase studies in superacids, Myhre and Yannoni have been able to obtain NMR spectra of carbocations at very low temperatures (down to 5 K) in solid-state matrices of antimony pentafluoride. The techniques of field desorption (FD) and electrohydrodynamic ionization (EHD) differ from SIMS, LD, and FAB in their physical basis and in features seen in the spectra. For example, the diquaternary ammonium salts discussed above yield intact doubly-charged ions, and fragmentation is less extensive. Nonetheless, the three classes of ions described for molecular SIMS are generally seen in FD (50). An EHD study of a series of diquaternary ammonium salts (51) led to the conclusion that the amount of internal energy deposited in EHD is less than in SIMS and FAB. For a few tissue samples, the matrix deposition can be performed manually. A matrix solution is sprayed onto the tissue section with a hand-held thin layer chromatography (TLC) sprayer or an artist airbrush. The reproducibility of manual matrix deposition is an issue. When the manual sprayer is used, the MALDI target plate with the tissue section is held vertically about 15-25 cm from the sprayer nozzle. It is recommended to spray multiple coats of matrix across the tissue section and each coating cycle consists of passing the sprayer two to hve times across the tissue section and allowing the tissue to dry for about 1-5 min. This process is usually repeated between 10 and 20 cycles. [Pg.407]
