Unlike the previously discussed compounds, which inhibit MTP in both liver and intestine, an intestine-selective orally-active MTP inhibitor JTT-130 (structure not yet disclosed) has been reported to decrease plasma cholesterol and TG in guinea pigs with no hepatic lipid accumulation [16]. Although further studies in human are needed, inhibitors that selectively target intestinal MTP might be a safer alternative as a treatment for hyperlipidemia than the liver-targeting MTP inhibitors. [Pg.164]

Since the structure of the liver is completely changed during fibrosis, distribution of a liver targeting compound has to be tested in the pathological state as well. Parts of the liver may become inaccessible by portosystemic shunts and individual cells may be hampered in the uptake of compounds by the excess extracellular matrix deposited around them. Uptake processes themselves may also be impaired. The phagocytic activity of KCs has been reported to be depressed in some forms of fibrosis [153,154] and some receptors, such as the... [Pg.106]

Quinoxaline-1,4-dioxides are synthetic antimicrobial agents the best-known members are carbadox and olaquindox. Both compounds are rapidly metabolized to monoxy- and desoxy- compounds. The final product from carbadox and the one most often determined, mainly in liver (target tissue), is quinoxaline-2-carboxylic acid. Carbadox and olaquindox are light-sensitive compounds and sample manipulations should be performed only under the minimum of indirect incandescent illumination. Carbadox and desoxycarbadox are insoluble in water but are soluble in chloroform and methanol, while olaquindox is slightly soluble in water and some organic solvents. The solubility, however, of quinoxaline-2-carboxylic acid can be easily monitored by adjusting the pH because it is a strong carboxylic acid (pK 2.88). [Pg.1049]

Storage sites for the tocopherols in the body include muscle and adipose tissues and the liver. Target tissues include the adrenals, pituitary, kidney, genital organs, muscles, liver, lungs, and bone marrow. [Pg.1706]
