Lung homogenates

Bead-bound PF-4540124 (2) was incubated with the soluble fraction of a mouse lung homogenate in the presence and absence of 1 (100 J.M). The type of protein captured was restricted by having 1 mM ADP and GDP in both samples. The beads were pelleted and washed before being treated (both samples) with 1 (100 iM) to elute specifically bound proteins. 

Holland and coworkers mentioned the TLC detection of a trihydroxy compound related to the hatching factor when homogenates of B. balanoides were incubated with 14C-AA, but no experimental details were given [143]. In contradistinction to the decrease in lysosomal enzyme activity observed in alveolar macrophages and bronchial lavage fluid, Dillard et al. reported that continuous ozone exposure (0.70-0.79 ppm for 5-7 days) resulted in an increase in the activity of some lysosomal hydrolases in rat whole-lung homogenates and lung fractions, including the soluble supernatant. The tocopherol concentrations of the diet had no effect on the findings.

Homogenize the lungs and perform 10-fold serial dilutions of lung homogenate in sterile PBS. 

Hyperoxia (48-hr exposure to >99% 02) decreased TAC of rat lung homogenates (cytosol fraction) by 26%. TAC of the acid-soluble fraction was even more depressed (by 40%). In our attempts to determine the possible role of free radical lipid peroxidation in smoke induced injury, the levels of lipid peroxidation products - thiobarbituric acid reactants, mainly malondialdehyde - wereQmeasured in lung homogenates with or without prior incubation at 37 C for one hour, contrary to our expectation, the levels of thiobarbituric acid reactants were found to be decreased, rather than increased, in the lungs of cigarette-smoke-exposed rats (Table III). Such a depression effect, however, was observed only when animals were exposed to whole smoke, and not to the gaseous phase of smoke. 

Sample preparation Lung. Homogenize (Ultra-Turrax T25) mouse lung in 1-3 mL pH 6.8 Soerensen phosphate buffer, centrifuge.
