Poly-L-lysine has been linked to specific peptides that bind the serpin enzyme complex receptor in an attempt to increase luminal airway epithehal gene transfer efficiency (28). The poly-L-lysine polymer has also been modified to enhance gene transfer efficiency. Kollen et al. (29,30) have demonstrated that lactosylated poly-L-lysine can significantly enhance gene DNA transfer to cultured CF cells in vitro.

Ligand-polylysiiie-DNA complexes for receptor-mediated gene transfer, for example, lactosylated poly-L-lysine has been shown efficiently to transfer CFTR cDNA to CF airway epithelial cells in vitro (40). 

Oishi et al. coupled lactosylated PEG to siRNA via an acid labile (S-thiopropio-nate bond, followed by complexation with poly(L-lysine) into polyion complex micelles. The complex was internalized by receptor-mediated endocytosis into hepatoma cells and the gene silencing effect of RNAi was considerably enhanced compared to the free conjugate, demonstrating the effect of pH-sensitive PEGylation [56].
