Stewart and Ballou have observed a great variation in the phosphorus content of diflFerent mannans, the highest being in Kloeckera brevis mannan, which has one phosphorus atom for every nine D-mannose residues. The species most commonly employed for thiamine determination are some Lactobacilli such as L.fermentum and L. viridescens (ATCC 12706). The latter is the microorganism most widely used to measure thiamine concentrations. It requires the intact thiamine molecule for growth. Other organisms such as Phycomyces blakeslekanus, Kloeckera brevis (ATCC 9774), Ochromonas danica and Neurospora crassa are available, but they are less useful. [Pg.288]

Strictly specific for thiamine are the yeast Kloeckera brevis and the bacteria Lactobacillus fermentum 36 and Lactobacillus viridescens. The first will measure 0.2-2.0 m g of thiamine, the two latter 5-40 m/ g. These methods were the first developed for biotin analysis and are still accepted as official methods because of their high sensitivity (nanogram level).
