Nutritional and cultural techniques for growing Ochromonas danica in light and darkness have been described (A2, H18). It was grown under constant illumination from five 40-watt warm-white fluorescent lights, ca. 1.0 m from the cultures. Transfers were made weekly in 10 ml of maintenance medium 1ml was used for transfer (Table 6).

Ochromonas danica Is reported to contain higher levels of lipids compared to Skeletonema (12). It is a freshwater species, and its nutritional requirements are richer than those of Skeletonema. 

Ochromonas danica culture (code LI298) was obtained from the culture collection of the University of Texas at Austin. The cells were cultured in a modified Hoaglands medium (11) for 10 days. For both species, the culture conditions were T - 22°C pH 8.2 light intensity 100 /jEinstein/m -sec light/dark cycle of cool-white fluorescence light 16/8hr COj source bubbling with filter sterilized air.

Mayers and Haines (1967) have isolated a sulfolipid, 1,14-docosyl disulfate, from the phytoflagellate, Ochromonas danica. Acid hydrolysis of the sulfatide yielded a diol. The structures of the sulfatide (lipoid sulfate ester) and the diol were determined by infrared, NMR, and mass spectroscopy. The infrared spectrum of the sulfatide diol had bands indicative of a long-chain diol 1060cm (primary alcohol CO), 1120 (secondary alcohol CO), and 718 cm (long-chain aliphatic CH2rocking). 

Chlorosulpholipids are found in some fungi (e.g. caldariomycin (CICH2COCH2CH2COOH) and certain algae. Ochromonas danica contains particularly high amounts of chlorosulpholipids where they represent almost half of the total membrane lipids.
