Enzymes can exist as two enantiomers, although only one enantiomer is found in nature. In 1992, Stephen Kent and co-workers reported the synthesis of both enantiomers of an enzyme that cuts peptide substrates, and showed for the first time that each enzyme acts only on the corresponding enantiomeric peptide substrate. [Pg.190]

The editors welcome the new contributors to the eleventh edition Doug Henry. Phillip Bowen, Stephen J. Cutler. T. Kent Walsh, Philip Proteau. and Michael J. Deimling. The editors extend thanks to all of the authors who have cooperated in the preparation of the current edition. Collectively, the authors rcprc.sent many years of teaching and rc.search experience in medicinal chemistry. Their chapters include summaries of current research trends that lead the reader to the original literature. Documentation and references continue to be an important feature of the book. [Pg.999]

In 1993, Stephen B. H. Kent (1945- ) and collaborators used automated peptide synthesis to construct all-D-amino acid superoxide dis-mutase. As expected it folded so as to be the mirror image of the native (all-L) enzyme. This D-protein is equally effective as the native protein in catalyzing the disappearance of superoxide (O2 ), a toxic species, because superoxide is achiral. The potential advantage of this approach is that D-proteins should not be destroyed by natural proteases nor should they ehcit immune responses. [Pg.390]

Further support and interest in my work was shown by Dr. Brian Kent, Dr. Stephen Schneider, and Mr. Ed Utt from the U.S. Air Force. After completion of the development of the Periodic Method of Moments, the PMM code, the Hybrid radome, low RCS antennas, and more, the funding from the Air Force shifted into more hardware-oriented programs. Fortunately, the U.S. Navy needed our help in designing very broadbanded bandstop panels. Ultimately, this work resulted in the discovery of surface waves unique to finite periodic structures, which are treated in great detail in this book. The help and advice from Mr. Jim Logan, Dr. John Meloling, and Dr. John Rockway is deeply appreciated. [Pg.377]

As an interesting illustration of this enzyme chirality principle, Stephen B. Kent, then at The Scripps Research Institute in La JoUa, CaMomia, synthesized the enantiomer of a natural enzyme by using the enantiomers of the amino acids normally found in proteins as building blocks. As expected, this synthetic enzyme enantiomer only catalyzed a reaction with a substrate that was the enantiomer of the chiral substrate utilized by the natural enzyme. [Pg.174]
