Of this product, 4.5 g in 30 cc of dry isopropyl alcohol are refluxed for 16 hours with 2.5 g of (3-chloroethyl dimethyl amine. The solution is cooled and filtered clear from the solid by-product. The solvent is removed under reduced pressure on the steam bath and the residue is washed with anhydrous ether. It is dissolved in ethyl acetate from which it crystallizes. It is the hydrochloride of (3-(dimethylamino)ethyl ester of 2-phenyl-2-( 1-hy-droxycyclopentyl) ethanoic acid, melting at 134° to 136°C. [Pg.413]

Preparation of 4-( -Chloroethyl)-3,3-Diphenyl-1Ethyl-2-Pyrrolidinone A solution of a,a -diphenyl-a -(1-ethyl-3-pyrrolidyl)-acetonitrile in 70% sulfuric acid was heated at 130°-140°C for 48 hours, poured onto ice, made basic with sodium hydroxide, and extracted with chloroform. The chloroform solution was acidified with hydrogen chloride gas, dried over sodium sulfate and concentrated. The residue was refluxed in 500 ml of thionyl chloride for 3 hours the resulting solution was concentrated in vacuo and the residue was crystallized from isopropyl ether. [Pg.537]

The residue is the 3-N-bis((3-chloroethyl)carbamate of estradiol. The compound melts at 101° to 103°C after recrystallization from isopropyl ether plus hexane (1 1). [Pg.579]

This residue is dissolved in isopropyl alcohol and 1 gram N-bis-chloroethyl-aniline is added to it. The mixture is refluxed for 3 hours. The solvent is removed at a reduced pressure, the residue is treated with 50% potassium carbonate, and extracted with ether. Methyl m-tolyl (8), ethyl m-tolyl, methyl n-butyl and methyl n-propyl sulfoxides were obtained in 100% e.e. This method was less successful when applied to methyl phenyl sulfoxide (5% e.e.) or to methyl isobutyl and methyl ethyl sulfoxides (25% e.e.). No complexes were formed between methyl o-tolyl, methyl p-tolyl, methyl 2-butyl and methyl isopropyl sulfoxides so these compounds could not be resolved using 7. A crystal structure of the 1 1 complex formed between 7 and 8 revealed that the partners were linked by OH—OS hydrogen bonds in endless zig-zag chains23. A clean and dry 20-gallon glass lined tank was charged with 19 L of water and 4.44 kg of sodium carbonate, after the carbonate had dissolved 4.29 kg (17.5 moles) of 5-(2-chloroethyl)-6-chloro-oxindole and 3.62 kg (16.5 moles) of 1-(l,2-benzisothiazol-3-yl)piperazine were added. The aqueous slurry was heated to reflux and the temperature maintained for 14 hours. When the reaction was complete the solution was cooled to 20°C and filtered. The wet product was reslurried in 23 L of isopropyl alcohol at room temperature for 2 hours. The product was collected by filtration on 2 large Buchner funnels, each was washed with 3.4 L of fresh isopropyl alcohol. The product was vacuum dried at 30° to 40°C. until no isopropyl alcohol remained, giving 5.89 kg (86.4% yield) of the desired free base which matched a standard sample by high performance liquid chromatography (HPLC). [Pg.3554]
