Insulator sequences

In other transcriptionally regulated Ad constructs described to date, the El A promoter has been removed and replaced with tumor or tissue-specific promoters (81-84). A list of different promoters that have been used to date is shown in Table 1. In some cases, interference from the nearby inverted terminal repeat (ITR) sequence can result in the loss of promoter strength and fidelity (85). Selectivity can be enhanced with the use of silencer (86) or insulator sequences (87). For example, the AdEla04i viras using the artificial a-fetoprotein (AFP) promoter to express ElA needed six copies of a silencer sequence to repress the promoter activity in normal hepatocytes. This virus showed specificity and potency for hepatoma carcinoma tumors and not in non-AFP- producing cells or in normal hepatocytes (86). [Pg.158]

A motor is able to sustain a negative sequence voltage up to 2% for short durations, while a sustained unbalance may deteriorate its insulation and affect... [Pg.276]

Hollomon s ethos, combined with his ferocious energy and determination, and his sustained determination to recruit only the best researchers to join his group, over the next 15 years led to a sequence of remarkable innovations related to materials, including man-made diamond, high-quality thermal insulation, a vacuum circuit-breaker, products based on etched particle tracks in irradiated solids, polycarbonate plastic and, particularly, the Lucalox alumina envelope for a metal-vapour lamp. (Of course many managers besides Hollomon were involved.) A brilliant, detailed account of these innovations and the arrangements that made them possible was later written by Guy Suits and his successor as director, Arthur Bueche (Suits and Bueche 1967). Some of these specific episodes will feature later in this book, but it helps to reinforce the points made here about Hollomon s coneeption of broad research on materials if I point out that the invention of translucent alumina tubes for lamps was... [Pg.9]

Since BamH I binds as a dimer to the palindromic sequence of 5 -GGATCC-3, two GG sites in the sequence should be equally insulated from one electron oxidation. In the absence of the protein, both G16G17 in ODN 35 (Fig. 8a, lane 2) and G8G9 in ODN 2 (Fig. 8b, lane 2) showed similar oxidization patterns under the irradiation conditions. In contrast, cleavage bands at both GG sites completely disappeared in the presence of BamH I (1.2 U/pL) (lane 3 in Fig. 8a,b). Simultaneous suppression of oxidation at both GG sites shows that insulation of both GG sites from one electron oxidation is due to the binding of BamH I to the recognition sequence. [Pg.186]

Electropolymerizable monomers that give rise to conducting electroactive polymers (CEPs) or insulating polymer thin films provide a convenient approach for the immobilization of DNA. More importantly, this method provides an easy means to achieve spatial separation of the ssDNA sequence... [Pg.180]

This topic is well covered by the contributions in this volume. CMP continues to be viewed as a surprisingly unique and flexible semiconductor fabrication technology by virtue of its ability to make manufactureable potential fabrication sequences that are either too cumbersome or too low in yield to be fabricated in any other manner. Using virtually any CMP polisher, a variety of materials of interest to IC fabricators can be planarized. These materials include insulators, semiconductors, interconnect metals, and barrier metallurgies. This means that once a user becomes adept in polishing one kind of material, typically oxide and W at first, other materials of interest and other semiconductor processing sequences become viable. [Pg.3]

The compound NCu02 (N = Ca0 86Sr014) is an insulator, but its structure, which is a simple defect perovskite made of layers (CuOa) sandwiched between layers (N), can be considered as the parent structure of a large family of superconductors. The sequence. ..(Cu0 )OiC(N)e o(Cu02)OjC. .. is in fact one of the building blocks of many compounds considered in this review. The refined parameters for NCu02 are given in Table 3. [Pg.201]

Despite the different assumptions in the two schemes described here, the properties of the calculated currents share the expected semiconducting features. This is basically due to the fact that they are formally acting on the r-stack in a similar way. Only further experimental investigation could shed more light on the discrimination of the gap-opening mechanism. Such an effort would eventually help in quantifying the influence of other insulating effects, which are possibly present in other DNA-based devices. They include (i) electron correlations and Coulomb blockade, (ii) localization effects due to the sequence variability, and (iii) local defects. [Pg.222]
