Inoculating loop

The agar plates were inoculated on the whole surface from a culture of M. rammaniana frozen at —80 °C using a sterile inoculation loop and incubated for 4 days at 28 °C. 

The five flasks with each 100 mL of medium (precultures) were inoculated with mycelium of M. rammaniana from the agar plates using a sterile inoculation loop and incubated on a laboratory shaker with 5 cm agitation radius at 28 °C and 220 rpm for 3 days. 

Sterile disposable Petri dishes and sterile inoculating loops for bacterial culture. 

Autoclave-sterilized flat end toothpicks or metal inoculating loop. 

Flame sterilize any instruments (e.g., inoculating loop) used for transferring your culture before and after transfer. 

With your sterile inoculating loop or the flat end of a toothpick, remove a very small drop of yeast and place it at the top of the agar medium. (If yeast is in a glycerol medium, allow the drop to melt on the medium.)... 

Pipettes, inoculating loops, petri dishes, agar, and other materials (including pre-mixed media) are found at many scientific supply houses or can be ordered from Difco Laboratories, Inc., Detroit, Michigan 48201. 

A simple technique can be used to physically separate spores so that individual dikaryotic (or even monokaryotic) cultures can be isolated in one step. The necessary equipment includes a bacterial (small) inoculating loop, several screw-cap vials of 20-30 ml, capacity, a flame and several sterile pipettes or small syringes. 

Bacterial binding to the sugars on the shdes could be removed specifically, so that the bound microbes can be apphed for further testing. Therefore, microarrays were incubated with E. coli cells and unbound cells were washed away. An inoculation loop was scraped over the sugar spots and streaked onto LB plates. After growth of the plates at 37 °C, E. coli cells could be detected on plates streaked with inoculation loops... 

Using an inoculating loop, remove a sufficient number of colonies from a culture plate that has been properly incubated for no longer than 24 h. Prepare a bacterial suspension equal to a 1 MacFarland standard see Note 4) in sterile saline and vortex. 

Incubator, steam bath, vibrator, pressure-filtering device for membrane filters, membrane-filters pore size 0.2 pm (1 pm = 0.001 mm), photometer with nephelometer attachment, filter Hg 436 nm, cuvettes (path length 10 mm), electric pH gauge, culture tubes with caps, Erlenmeyer flasks (300 ml), graduated pipettes (1 ml), graduated pipettes (10 ml), measuring cylinders (100 ml), glass beads (diameter approx. 2 mm), inoculation loop, aqueous sodium hydroxide, hydrochloric acid.

Reshape loop portion of inoculating loop into a pick. Flame sterilize, cool, and collect a portion of mycelial mass and inoculate corners of the agar square. 

Included here are necessary supplies such as microscope slides and inoculating loops. A package of 12 general-purpose inoculating loops costs 55. Calibrated inoculating loops cost from 40 to 50 for l- iL and 100 for lO-pL delivery volumes. The latter are used for cell-counting work. Microscope slides cost from 16 to 24 per gross. 

Inoculate Agrobacterium from Petri dish into 10 ml LB (with appropriate antibiotics) using a sterile inoculating loop.

Day-1 Take a small amount (2-3 colonies) of the fireshly grown yeast cells with an inoculation loop and use to inoculate 5 mL of liquid YES medium in a test tube or small Erlenmeyer flask for overnight cultures (Fig. 1).
