Infrared individual cell

Several additional instrumental techniques have also been developed for bacterial characterization. Capillary electrophoresis of bacteria, which requires little sample preparation,42 is possible because most bacteria act as colloidal particles in suspension and can be separated by their electrical charge. Capillary electrophoresis provides information that may be useful for identification. Flow cytometry also can be used to identify and separate individual cells in a mixture.11,42 Infrared spectroscopy has been used to characterize bacteria caught on transparent filters.113 Fourier-transform infrared (FTIR) spectroscopy, with linear discriminant analysis and artificial neural networks, has been adapted for identifying foodbome bacteria25,113 and pathogenic bacteria in the blood.5... [Pg.12]

Vibrational spectroscopy provides for the analysis of the chemical composition, molecular structures, conformations, and interactions in a sample. Two methods that are commonly used for molecular vibrational analysis are infrared (IR) and Raman spectroscopy. One of the limitations of the former, which relies on measuring the absorption or reflection of mid-IR radiation by molecular bonds, is the low spatial resolution afforded by the technique, allowing only chemical information from a large group of cells to be obtained. Raman spectroscopy, on the other hand, offers high spatial resolution subcellular chemical analysis of an individual cell. It relies on the scattering of photons by molecular bonds, which yields chemical information about the molecular stmcture and conformations in the sample. It has seen an increase in its use for biological studies because it offers many attractive features. The method is able to... [Pg.148]

A chapter entitled Infrared and Raman Spectroscopy and Spectral Imaging of Individual Cells was published in 2009 in the first edition of this volume [1]. In the ensuing 4 years, enormous progress was achieved in the field of vibrational spectroscopy of individual cells in fact, so much progress that in this revised chapter, only infrared (IR) results from the authors laboratory, the Laboratory of Spectral Diagnosis (LSpD), will be presented. For the Raman results from the LSpD, the reader is referred to the original literature [2-8]. [Pg.181]
