Hyperforin, the major constituent in Hypericum perforatum (St. John s Wort), inhibits the enzymatic activity of 5-lipoxygenase and COX-1 in platelets, acts as a dual inhibitor of 5-lipoxygenase and COX-1, and might have some potential in inflammatory and allergic diseases connected to eicosanoids (32), Several Hypericum species are of medicinal value in Asia and the Pacific. One of these is Hypericum erectum Thunb., the potential of which as a source of 5-lipoxygenase is given here. [Pg.41]

Several natural products have been evaluated in rodent models of nicotine withdrawal. An extract of Hypericum perforatum (St. John s Wort, a putative antidepressant, and inhibitor of serotonin reuptake) reversed somatically expressed withdrawal behaviors and locomotor depression in spontaneous withdrawal (Catania et al. 2003). A benzoflavone compound isolated from Passiflora incarnata, interfered with the induction of physical dependence. Coadministration with chronic nicotine prevented various subsequent indicators of withdrawal syndrome in the mouse, including jumping, locomotor inactivity, immobility in the swim test and naloxone-precipitated escape jumping (Dhawan et al. 2002). [Pg.425]

Several species of plant also contain compounds that have been shown to act as agonists on benzodiazepine receptors. These include Valeriana officinalis which contains hydroxypinoresinol, Matricaria recutita which contains 5,7,4 -trihydroxyflavone, Passiflora coeruleus which contains chrysin and Karmelitter Geist which contains amentoflavin. Hypericum perforatum (St John s Wort) also contains unknown compounds which have affinity for... [Pg.451]

Sigma receptor ligands bind to metabotropic GPCRs as well as ionotropic cr-Rs (Chapter 3). Endogenous ligands for cr-Rs include some opiates. Sigma-R activation can have antitussive, anxiolytic and ulceroprotective effects. Hypericin from Hypericum perforatum (St John s wort) is... [Pg.167]

Hypericum perforatum (St. John s wort), which is used for mild depression, is an inducer of cytochrome P-450. It caused a rapid dramatic fall in ciclosporin blood concentrations, resulting in acute heart transplant rejection in two patients aged 61 and 63 years (262). It produces an average 50% reduction in ciclosporin blood concentrations (263). At least three other case reports have clearly confirmed that St. John s wort is dangerous in transplant patients, because it produced a rapid and dramatic reduction in blood ciclosporin concentration and resulted in acute organ rejection in two patients (264—266). Induction of CYP3A4 and/or P-glycoprotein was the most likely mechanism. [Pg.759]
