If the TM domain crosses the bilayer in the N-terminal to C-terminal direction, the CRAC domain will probably interact with a cholesterol molecule located in the cytoplasmic leaflet of the membrane. This is the case for single-pass type I membrane proteins and for TM domains 1,3,5, and 7 of G-proteins coupled receptor (GPCR) proteins with seven TM domains. A representative example of this type of interaction is given by the fifth TM domain of the human type 3 somatostatin receptor. An interaction of the CRACI domain with cholesterol in the exofacial leaflet is possible if the TM domain crosses the bilayer in the C-terminal to N-terminal direction, as is the case for single-pass type II membrane proteins and for TM domains 2, 4, and 6 of GPCR proteins. [Pg.141]
