Anti-tumor ether lipids are stmctural analogs of PAF but lack the readily hydrolysable ester substituent at the sn-2 position of the glycerol moiety. They contain a long carbon chain at sn-1 and a short chain at the sn-2 position. At the sn-3 position, phosphocholine is present as the head group. Examples of antitumor ether lipids are Edelfosine (l-O-octadecyl-2-O-methyl-rac-glycero-3-phosphocholine, Et-lS-OCHj), Ilmofosine, and Miltefosine (hexadecyl phosphocholine, HePC). They are unnatural compounds that are metabolized by mammalian tissues at a low rate (Eigure 1). [Pg.119]

Very recently, the receptor-independent cytotoxic effects of synthetic 1-0-hexadecyl-2-azelaoyl-sn-glycero-3-phosphocholine (HAz-PC) were studied. This compound is an ether analogue of l-hexadecanoyl-2-azelaoyl-sn-glycero-... [Pg.360]
