Contamination of the production vessel leads to serious financial penalties and each step in the inoculum train is monitored for contamination. To reduce the risk of contamination during sampling it is usual to take a sample from the residue left in each vessel after its contents have been transferred to the next reactor. Since these contamination checks are retrospective, a heavy reliance is placed on the growth characteristics of the production organism. Kinetic variables such as growth rate and oxygen consumption rate are also used to assess the quality of the inoculum. [Pg.206]

Growth characteristics of cells exposed to water stress mimic some of the structural responses of organised plant tissues. A frequently observed response of plants exposed to water stress is a reduction in cell size (Cutler, Rains Loomis, 1977). This cellular phenomenon was observed in tomato cells stressed with PEG (Handa et al., 1983). Concomitantly with a decrease in cell size with increasing osmotic stress was a reduction in fresh weight. In contrast the dry weight was not affected. [Pg.183]

A methanogenic bacterium was isolated from oil reservoir brines by enrichment with trimethylamine. Methane production occurred only with trimethyl-amine compounds or methanol as substrates. Sodium ions, magnesium ions, and potassium ions were all required for growth. This organism appears to be a member of the genus Methanohalophilus based on substrate utilization and general growth characteristics [695]. [Pg.223]

Ex vivo procedures involve removing cells and transfecting or transducing them. The cells should be checked to confirm that they are all healthy and that they are still expressing their normal surface markers, etc observations of normal growth characteristics can also be reassuring. Animal studies are of limited value to test the safety of transfected or transduced human cells. [Pg.423]

The HCE-T model was established in 1995 by Araki-Sasaki and coworkers [56], It was formed by transfection of human corneal epithelial cells from a 47-year-old female donor with a recombinant SV40-adenovirus, which lacked the origin of viral replication (i.e., not able to produce viral particles). The cell line has good growth characteristics and shows a cobblestone-like appearance. The cell line is intensively used by Urtti and coworkers and represents a standard... [Pg.292]
