Glucosidase cloning

In this article, we report the cloning in Escherichia coli and sequencing of an Orpinomyces P-glucosidase cDNA. The enzyme was overexpressed in and secreted from the yeast Saccharomyces cerevisiae. Physiochemical properties of the secreted enzyme were determined after it was purified. Its applicability for cellulose saccharification was also assessed.

The glucoalkaloid raucaffricine has been reported to be synthesized in Rau-volfia cell cultures as the major alkaloid. Isolation and cloning of the cDNA of raucaffricine glucosidase enabled overexpression of the recombinant enzyme which can be prepared in applicable amounts and used efficiently in chemo-enzymatic routes.
