Gestation

Recently, there has been increasing interest in the development of test methods to assess the possible adverse functional effects of exposing the fetus, both early and late in gestation (112,113). 

Studies in which pregnant rats and mice were exposed to 1250 ppm of methylene chloride for seven hours a day on days 6—15 of gestation indicated no significant maternal, embryonal, or fetal toxicity (34). Methylene chloride was shown to be nonteratogenetic to either animal at the concentration studied.

The expression of all three PPAR isotypes peaks in the rat central nervous system between days 13.5-18.5 of gestation, and while expression of both PPARa and PPARy decline post-natally, expression of PPARS remains high (except for the retina, where all three isoforms are expressed in the adult rodent). An important role for PPARS in CNS development is underscored by the occurrence of defective myelination in the PPARS-null mouse.

After administration of a single oral dose of dibutyltin diacetate of 22 mg/kg body weight to pregnant rats on day 8 of gestation, both dibutyltin and monobutyltin were detected in the embryos, indicating placental transfer (Noda et al., 1994). Nakamura et al. (1993) also detected dibutyltin in embryos after dosing the mother orally on days 7-17 of gestation.
