Free light chains

The presence of oligoclonal free kappa and lambda chains in CSF is a sensitive indication for recent antigenic immune response within the central nervous system, comparable with IgM. The detection of oligoclonal free kappa chains in CSF supports the diagnosis of multiple sclerosis. In addition, free light chains can also be found in the CSF of patients having inflammatory diseases of the central nervous system (LI). [Pg.32]

Purification of humanized monoclonal antibodies has also been published using nickel affinity chromatography.190 Directly after filtration of feed stock, antibodies were adsorbed and elution was achieved by a descending pH gradient from 7.5 to 4.25. Purity obtained was about 90% and the fraction did not contain free albumin nor free light chains. Recovery was also reported as greater than 9Q% from ascitic fluids as well as from cell culture supernatants. [Pg.593]

In monoclonal gammopathies, the IFE patterns usually yield a distinct, sharply defined precipitin band with one heavy-chain and one light-chain antiserum. These bands match the location of the particular immunoglobulin in the reference pattern (Figure 20-10). A second, fainter band of free light chains may also be present. [Pg.586]

If a monoclonal band, or free light chains or heavy chains, are detected by IFBP, are they the pathogenic ones These abnormalities demonstrate a dysimmune milieu within the patient and a suspicion that the detected abnormality might be the cytotoxic one. However, conceptually the actual dysimmune cytotoxicity could be, qualitatively, due to a different monoclonal nano-farmly that is not quantitatively high enough above the detection threshold to be identified as "monoclonal."... [Pg.43]

Chlorination of Methane. Methane can be chlorinated thermally, photochemicaHy, or catalyticaHy. Thermal chlorination, the most difficult method, may be carried out in the absence of light or catalysts. It is a free-radical chain reaction limited by the presence of oxygen and other free-radical inhibitors. The first step in the reaction is the thermal dissociation of the chlorine molecules for which the activation energy is about 84 kj/mol (20 kcal/mol), which is 33 kJ (8 kcal) higher than for catalytic chlorination. This dissociation occurs sufficiendy rapidly in the 400 to 500°C temperature range. The chlorine atoms react with methane to form hydrogen chloride and a methyl radical. The methyl radical in turn reacts with a chlorine molecule to form methyl chloride and another chlorine atom that can continue the reaction. The methane raw material may be natural gas, coke oven gas, or gas from petroleum refining. [Pg.514]

The regioselectivity of addition of Itydrogen bromide to alkenes can be complicated if a free-radical chain addition occurs in competition with the ionic addition. The free-radical reaction is readily initiated by peroxidic impurities or by light and leads to the anti-Markownikoff addition product. The mechanism of this reaction will be considered more fully in Chapter 12. Conditions that minimize the competing radical addition include use of high-purity alkene and solvent, exclusion of light, and addition of free-radical inhibitors. ... [Pg.353]

In order to induce the free-radical chain reaction, a starter compound such as dibenzoyl diperoxide, azo-Zj -(isobutyronitrile) or tcrt-butyl hydroperoxide or UV-light is used. The commercially available, technical grade N-bromosuccinimide contains traces of bromine, and therefore is of slight red-brown color. Since a small amount of elemental bromine is necessary for the radical... [Pg.300]

Dimethacrylate monomers were polymerized by free radical chain reactions to yield crosslinked networks which have dental applications. These networks may resemble ones formed by stepwise polymerization reactions, in having a microstructure in which crosslinked particles are embedded in a much more lightly crosslinked matrix. Consistently, polydimethacrylates were found to have very low values of Tg by reference to changes in modulus of elasticity determined by dynamic mechanical analysis. [Pg.427]

The specific labeled separated protein fractions blotted on a nitrocellulose membrane or specific immunoflxation-separated protein fractions in polyacrylamide after isoelectric focusing make it possible to detect some additional bands in CSF, i.e., IgM, IgA, free kappa or lambda light chains of specific antibodies (i.e., antiherpes, anti-borrelia, or anti-HIV) (LI, M3). [Pg.31]
