Microbial mats have been a longstanding focus of N2 fixation studies (Paerl et al., 1996, 2000) (Table 4.1). Most recently, appHed molecular approaches have revealed much more information about these systems. Zehr et al. (1995) examined a microbial mat from an intertidal mat from coastal North Carolina and found a wide variety of diazotrophic bacteria were present, with the majority of sequences related to niJH sequences of anaerobes (sulfate reducers and Clostridia), particularly deeper in the mat.

The BASF representatives were chemist Carl Bosch and catalyst expert Alwin Mittasch. They were there to find out whether the backing for Haber s nitrogen fixation studies by the head of the managing board, Heinrich von Brunck, had finally paid of Though the apparatus had been tested and found successful, the initial outcome of the demonstration seemed disappointing the apparatus refused to work. Carl Bosch left to attend to other business. Mittasch remained in the laboratory and later in the day was rewarded when the apparatus delivered 100 cubic centimetres of ammonia. He was convinced Haber had overcome the laboratory difficulties involved in the synthesis of ammonia. Patents for the continuous process were filed in Germany and elsewhere, and Haber came to an agreement with BASF over royalties. ... 

Except for some of the fixation studies (for which Raman data were also acquired) and for studies in aqueous media (see later), all sample preparations used reflection-coated glass slides as sample substrates. Raman data are prone to show some glass signatures when these slides are used consequently, some studies that relied on a combination of Raman and IR data were carried out on CaF2 disks. Similarly, CaFj was used as a substrate for IR studies of live cells, as prolonged exposure to water damaged the reflective slides. 

Aside from the fixation studies, which were carried out for both exfoliated and cultured cells, the majority of the work presented in the sections above has dealt with exfoliated cells that is, it reported a truly new form of cytology, namely SCP. To the best of our knowledge, work on exfoliated cells at the cell-by-cell level is now being carried out exclusively at the LSpD, and the size of the data sets at the LSpD far exceeds all previously collected data sets combined [35,44]. At the time of writing this summary, it appears that the SCP has matured to a level that allows for detection of cellular abnormalities, such as dysplasia, cancer, and viral infection in exfoliated cells and, thus, is poised to be applied to areas where classical cytology has very poor performance, in many cases below 50% accuracy.
